生血宝合剂对心梗后心衰大鼠心功能及心肌结构的影响

摘要:
目的 观察生血宝合剂对心梗后心衰大鼠心功能及心肌结构的影响。 方法 以冠脉结扎配合力竭式游泳制作心梗后大鼠心衰模型,将造模成功存活的大鼠随机分为模型组、赖诺普利组、生血宝合剂组,每组10只;未行冠脉结扎术的大鼠10只为对照组。各组均通过灌胃给药或蒸馏水,给药6周后超声心动图检测左室收缩末期容积(ESV)、左室舒张末期容积(EDV)、左室射血分数(LVEF)和左室每搏输出量(SV),后处死全部大鼠,取心肌组织HE染色观察心肌组织结构变化。 结果 与对照组比较,模型组、生血宝合剂组及赖诺普利组ESV、EDV明显升高,LVEF、SV显著下降,差异均有显著性意义,P<0.05。与模型组比较,生血宝合剂组及赖诺普利组ESV、EDV显著下降,LVEF、SV明显升高,差异均有显著性意义,P<0.05。生血宝合剂组与赖诺普利组比较,ESV、EDV、LVEF及SV数值,差异均无显著性意义,P>0.05。HE染色显示,对照组心肌细胞排列整齐、致密、结构清晰,细胞外间质较少,可见少量成纤维细胞。模型组心肌细胞淡染,排列紊乱,细胞间隙增大、形态模糊,成纤维细胞增多,伴有炎症细胞浸润。生血宝合剂组及赖诺普利组心肌细胞形态较清晰,成纤维细胞和炎症细胞少见。 结论 生血宝合剂可改善心梗后心衰大鼠心功能,修复受损的心肌组织,起到治疗心衰的作用。

Abstract:
Objective To observing Shengxuebao mixture effect on cardiac function and cardiac structure for heart failure after myocardial infarction rats. Methods coronary artery ligation method and swimming exhaustion caused by the rats heart failure model after myocardial infarction,the Survivals were randomly divided into model group,Lisinopril group,Shengxuebao mixture group,no coronary artery ligation in rats as control group,10 rats in each group,by gavage administration or distilled water.After 6 weeks of medication,by doppler echocardiography machine detected between groups in the rats cardiac function;Using HE staining to observe the structure changes of myocardial tissue. Results Compared with control group, model group, Shengxuebao mixture group and lisinopril group ESV and EDV were significantly higher, LVEF and SV dropped significantly, P<0.05.Compared with model group, Shengxuebao mixture group and lisinopril group ESV, EDV dropped significantly, LVEF and SV increased significantly, P<0.05.Shengxuebao mixture group Compared with Lisinopril group,ESV,EDV,LVEF and SV were no significant difference,P>0.05.HE staining results: Control group myocardial cell neatly, compact, the structure was clear, the outer cell mass was less, can be found in a small amount of fibroblasts.Model group myocardial cell light dye, disordered arrangement, intercellular space increases, ambiguous, fibroblasts increased, accompanied by inflammatory cells infiltration.Shengxuebao mixture group and lisinopril group myocardial cell morphology was clear, that can be found small amount of fibroblast cells and inflammatory cells. Conclusion Shengxuebao mixture can improve cardiac function in rats with heart failure after myocardial infarction and repair damaged heart tissue, thus achieve effect for the treatment of heart failure.
