目的 研究中间型滋养细胞的凋亡及过氧化物酶体增殖物激活受体γ(PPARγ)在子痫前期患者胎盘中的表达及临床意义。 方法 收集2014-2016年间大连市妇女儿童医疗中心住院分娩的正常妊娠晚期(正常组)、轻度子痫前期(轻度子痫前期组)及重度子痫前期(重度子痫前期组)产妇胎盘组织,各30例。采用 TUNEL法检测3组胎盘底板中间型滋养细胞的凋亡并计算凋亡指数(AI),采用免疫组化S-P法检测PPARγ的表达情况。 结果 正常妊娠晚期组、轻度子痫前期组及重度子痫前期组胎盘滋养细胞的AI分别为1.5860±0.2541、5.2609±2.8643及9.0000±6.2763,呈增高趋势,各组间差异有显著性意义(P<0.05)。PPARγ在正常妊娠晚期组、轻度子痫前期组及重度子痫前期组的表达阳性率分别为46.7%,93.3%,100%,呈增强趋势,各组间差异有显著性意义(P<0.05)。子痫前期胎盘组织中PPARγ与凋亡细胞的表达呈正相关(r0.591, P<0.05)。 结论 PPARγ可能通过诱导胎盘滋养细胞的凋亡参与子痫前期的发病。 |

Objective To investigate the clinical significance of apoptosis index (AI) and PPARγ expression in the placenta of patients with preeclampsia. Methods The placenta tissues from normal-term pregnancy, light preeclampsia and severe preeclampsia patients (30 cases in each group) were collected from 2014-2016 years in Dalian Municipal Women and Children,s Medical Center. TUNEL was performed to measure AI and immunohistochemistry (S-P method) was used to detect PPARγ expression in intermediate trophoblast of placental floor. Results AI was 1.5860±0.2541, 5.2609±2.8643 and 9.0000±6.2763 in normal-term pregnancy group, the light preeclampsia group and the severe preeclampsia group, respectively (P<0.05). The PPARγ expression frequency was 46.7%, 93.3% and 100% in normal-term pregnancy group, the light preeclampsia group and the severe preeclampsia group, respectively (P<0.05). The AI was positively correlated with PPARγ expression in placenta of patients with preeclampsia (P<0.05). Conclusion PPARγ could involve in the pathogenesis of preeclampsia by inducing apoptosis of placental trophoblast cells. |
