半边莲煎剂对肝癌的抑制作用及对P27和BCL-2表达的影响

摘要:

目的　研究半边莲煎剂对肝癌H22荷瘤小鼠的抗肿瘤作用。　方法　建立肝癌H22荷瘤小鼠实体瘤模型，将24只小鼠分为3组，生理盐水组、半边莲煎剂组和5-氟脲嘧啶组，每组各8只。通过测定荷瘤小鼠的瘤重、抑瘤率观察半边莲煎剂对肿瘤生长的抑制作用，并采用免疫组化技术检测肿瘤组织中P27和BCL-2的表达情况。　结果　半边莲煎剂组瘤重与生理盐水组相比明显减小，差异有显著性意义（P<0.05）。半边莲有明显的抑瘤作用，其抑瘤率为39.48%。P27和BCL-2在3组中均呈阳性表达，半边莲煎剂组的P27表达增强，BCL-2表达减弱，与生理盐水组比较，差异均有显著性意义（P<0.05）；而半边莲煎剂组与5-氟脲嘧啶组比较，差异无显著性意义（P>0.05）。　结论　半边莲煎剂具有明显的抑瘤作用，其作用机制可能与上调P27和下调BCL-2表达有关。

Abstract:

Objective　To study the anti-tumor effects of Banbianlian on H22 hepatocelluar carcinoma-bear mice.　Methods　H22 hepatocelluar carcinoma-bear mice model was established; 24 mice were divided equally into three groups: normal saline treated group, Banbianlian treated group and 5-Fu treated group. The tumor was removed and measured. P27 and BCL-2 expression in the tumor tissues were examined in immunohistochemistry. 　Results　Banbianlian inhibited the tumor growth of hepatocelluar carcinoma-bear mice with the inhibitory rate 39.48％. The expression of P27 and BCL-2 in three groups were positive. P27 expression was stronger, while BCL-2 expression was weaker in Banbianlian treated group than those in normal saline treated group (P<0.05). Two protein expression were not significantly different between Banbianlian treated group and 5-Fu treated group (P>0.05).　Conclusion　 Banbianlian could inhibit tumor growth, which could be related to the abnormal expression of P27 and BCL-2.
