蓝莓花青素提取物对H2O2诱导人胃癌

摘要:
目的 在细胞水平上分析蓝莓花青素提取物的抗氧化活性。 方法 用6.25 μg/mL、12.5 μg/mL、25 μg/mL、50 μg/mL和100 μg/mL蓝莓花青素提取物预处理人胃癌BGC-823细胞24 h后,再加入1 mmol/L H2O2诱导损伤2 h。按不同处理分为对照组、H2O2处理组和蓝莓花青素保护组。采用MTT比色法检测细胞活力、相差显微镜观察细胞形态、荧光探针DCFH-DA测定细胞活性氧(ROS)生成量、比色法和可见光法测定谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)的活性,分析不同浓度的蓝莓花青素提取物预处理对H2O2诱导BGC-823细胞氧化损伤的影响。 结果 与H2O2处理组相比较,6.25 μg/mL、12.5 μg/mL、25 μg/mL、50 μg/mL和100 μg/mL蓝莓花青素提取物预处理均能增加BGC-823细胞活力(P<0.05)。25 μg/mL和100 μg/mL蓝莓花青素提取物预处理能阻止H2O2诱导的BGC-823细胞形态损伤,抑制ROS生成,增加GSH-Px和CAT的活性。 结论 蓝莓花青素提取物对H2O2诱导人胃癌BGC-823细胞氧化损伤具有保护作用。

Abstract:
Objective To explore the antioxidant activity of anthocyanins extract from blueberry on cells.  Methods The human gastric cancer BGC-823 cells were treated with anthocyanins extract from blueberry for 24 h with 6.25 μg/mL, 12.5 μg/mL, 25 μg/mL, 50 μg/mL and 100 μg/mL, then treated with H2O2 (1 mmol/L) for 2 h. The cell viability was measured by MTT assay, the cell morphological change observed by phase contrast microscope, reactive oxygen species (ROS) measured with the fluorescent probe 2′,7′-dichlorofluorescin diacetate (DCFH-DA).The activity of glutathione peroxidase (GSH-Px) and catalase (CAT)were analyzed by the chromatometry and the visible spectrophotometry.  Results Pretreatment with varied concentrations of anthocyanins extract from blueberry all increased BGC-823 cell viability (P<0.05). Pretreated with 25 μg/mL and 100 μg/mL anthocyanins extract inhibited the formation of ROS and elevated the activities of GSH-Px and CAT.  Conclusion The anthocyanins extract from blueberry could protect human gastric cancer BGC-823 cell to oxidative injury induced by H2O2.
