目的 探讨去势SD大鼠自体卵巢组织两种不同移植方法的效果比较。 方法 按照移植部位不同将SD大鼠随机分为肾被膜下移植组、颈部皮下移植组和对照组,每组20只。记录各组手术所用时间、移植后大鼠动情周期变化,并给予HMG促排卵48 h后回收卵巢组织进行HE染色观察组织学变化和计算卵子存活率。 结果 肾被膜下组较颈部皮下组所用手术时间短,分别为(18.8±2.5) min和(20.7±2.8)min,P<0.05;肾被膜下组与颈部皮下组移植后大鼠动情周期出现率分别为81.25%与61.11%,差异有显著性意义(P<0.05),大鼠恢复动情周期天数无差异。两组促排后大鼠移植部位卵巢组织回收率分别为:肾被膜下组81.3%,颈部皮下组55.6%,差异有显著性意义(P<0.05)。但两移植组回收卵巢组织卵泡存活数较对照组减少(均P<0.05),两移植组间差异无显著性意义(P>0.05)。 结论 与颈部皮下移植相比,大鼠卵巢组织自体肾被膜移植可获得更好的效果。 |

Objective To investigate the influence of two transplantation methods on auto-transplantation of mouse ovarian tissue in SD mice. Methods According to the operation methods,the mice were divided into three groups: kindey capsule transplantation group, neck subcutaneous transplantation group and control group(n=20). The operation time were recorded after operation.Vaginal smears to observe estrus cycle in mice,two weeks later giving HMG to stimulate ovarian oviposit, after 48 h recycling ovarian tissue and HE dyeing to observe histological changes and the follicle survival rate was recorded. Results Significantly lengthened operation time was observed in neck subcutaneous transplantation group than kindey capsule transplantation group[(20.7±2.8)min vs. (18.8±2.5)min,P<0.05]. Kindey capsule transplantation group and neck subcutaneous transplantation group mice appeared estrus rate were 81.25% and 61.11% respectively(P<0.05),there was no difference between the two transplantation groups back estrus days. The recovery rate of kindey capsule group was 81.3% after giving HMG, and the recovery rate of the neck subcutaneous group was 55.6%(P>0.05).The follicle survival rates in the two transplantation groups were significantly less than control group (P<0.05),while there was no significant difference between the two transplantation groups (P>0.05). Conclusion Kindey capsule is more suitable for auto-transplantation of mouse ovarian tissue than neck subcutaneous, auto-transplantation of mouse ovarian tissue kindey capsule can get better results. |
