安体舒通对糖尿病大鼠肾脏Nephrin蛋白表达的影响

摘要:
[摘要] 目的 探讨安体舒通对糖尿病(DM)大鼠肾脏Nephrin蛋白表达的影响。 方法 SD大鼠分为对照组、糖尿病肾病(DN)组、安体舒通组。腹腔注射链脲佐菌素(50 mg/kg)建立糖尿病大鼠模型,实验12周末检测肾功能、蛋白尿、脂蛋白,Western Blot检测足细胞Nephrin 蛋白含量(P<0.05)。 结果 较安体舒通组,DN组大鼠尿素氮(BUN)、血肌酐(Ccr)下降,24 h尿微量白蛋白排泄率(UAER)增多,脂蛋白升高,且肾脏Nephrin 蛋白表达下调,差异有显著性意义(P<0.05)。 结论 安体舒通对糖尿病肾病肾功能有保护作用,其机制可能与其上调Nephrin蛋白有关。

Effect of spironolactone on Protein Expression of Nephrin in kidney of Diabetic Nephropathy Rats

Abstract:
[Abstract] Objective To observe the effects of spironolactone on the expression of nephrin in the kidney of diabetic nephrophathy rats.  Methods Healthy male Sprague-Dawley(SD)rats were randomly divided into normal control (NC) and experimental groups.Rat models of diabetic nephropathy were established by intraperitoneal injection of streptozotocin; After the modeling, the experimental group were randomly divided into diabetic nephrophathy (DN) group and spironolactone group. At the end of 12th week, blood urea nitrogen(BUN), creatinine clearance rate(Ccr), 24h urine protein(24hUpro) and lipoprotein A were determined, the expression level of Nephrin protein was observed by western blot.  Results Compared with spironolactone groups, BUN, Ccr,24Upro and lipoprotein A in DN group statistically increased; the expression level of nephrin protein in spironolactone group was increased compared with DN group.  Conclusion Spironolactone can improve the renal function of rats with diabetic nephropathy. Its mechanism may be ralited to the increasing of nephrin expression.
