双光子显微镜(two-photon microscopy,TPLSM)具有三维成像功能,在分辨率和成像深度方面均具有明显优势。同时,它对样本的损伤程度也明显低于传统成像方法,这使双光子显微镜技术在神经科学研究中的地位日益提升。此外,TPLSM在光化学,尤其是光解释放笼状分子方面具有无可比拟的优势,该优势为进一步深入研究组织乃至细胞的细微结构、分子成分及细胞功能提供了一种可靠的方法。本文综述TPLSM的上述优点在脑片、整体脑结构样品和活体脑功能中的研究应用,期待双光子显微镜技术在神经科学研究中发挥更大的作用。 |

Two-photon laser scanning microscopy (TPLSM) has the function of three-dimensional imaging,and it has obvious progresses in resolution and imaging depth.Meanwhile,damages made by this imaging technology are much slighter compared to conventional imaging methods.These advantages advarce TPLSM technology more and more significant in neuroscience research.In addition,advantages of TPLSM in photochemical,especially in photolytic release of caged molecular,is unprecedented.TPLSM offers a reliable method for deep study of the fine structures,molecular components and cellular functions from tissue to single cells.This review summarized the applications of those advantages in the research of neuroscience,including studies of brain slices,whole isolated brain preparations and studies on living animals,so that TPLSM could play a more important role in neuroscience research. |
