缺氧诱导因子-1α及血管内皮生长因子在卵巢上皮性肿瘤中的表达

摘要:
[目的] 探讨缺氧诱导因子-1α(HIF-1α)及血管内皮生长因子(VEGF)在卵巢肿瘤组织中的表达。[方法] 选取卵巢癌组织58例、交界性肿瘤23例作为实验组,正常卵巢组织38例作为对照组。应用免疫组化SP法和蛋白印迹免疫分析法检测三者HIF-1α及VEGF表达。[结果] HIF-1α在卵巢癌、交界性肿瘤和正常卵巢阳性表达率分别为86.21%、47.83%和13.16%,三者比较差异均有显著性意义;HIF-1α的表达与临床分期正相关(P<0.05),且随病理分级增加而增强(P<0.05);VEGF在三种组织均有表达,其阳性表达率分别为98.28%、82.61%和57.89%,两两比较差异均有显著性意义(P<0.05);VEGF表达与卵巢癌临床分期正相关;且随病理分级增加而增强(P<0.05)。蛋白印迹免疫分析显示卵巢癌组织HIF-1α及VEGF蛋白表达量分别是正常卵巢组织的4.58±1.73及4.32±1.55倍。[结论] 检测卵巢癌组织中HIF-1α及VEGF有助于卵巢癌的早期诊断、预后判断。

Abstract:
[Objective] To investigate the expression of HIF-1α and VEGF in ovarian cancer tissue.[Methods] The expression of HIF-1α and VEGF were assessed by imunohistochemical staining and western-blot method in 58 cases of ovarian cancer, 23 cases of carcinoma in situ and 38 cases of nomal ovarian tissue. [Rusults] Immunohistochemical staining:HIF-1α level in ovarian carcinoma was much higer than those in carcinoma in situ and nomal tissue, their positive staining rate were 86.21%, 47.83% and 13.16% (P<0.05). The expression of HIF-1α elevatated as the clinical stage increased (P<0.05) and the pathologic grade increased (P<0.05). VEGF level in ovarian carcinoma was higer than those in carcinoma in situ and nomal tissue, their positive staining rate was 98.28%, 82.61%, and 57.89% (P<0.05). The expression of VEGF elevatated as the clinical stage increased (P<0.05) and the pathologic grade increased (P<0.05). VEGF has positive correlation with HIF-1α. Western-blot: the protein band of HIF-1α and VEGF is at 120 kD and 23 kD, the OD of HIF-1α is 4.58±1.73 times while VEGF is 4.32±1.55 times in early invasive stage as that in normal tissue. [Conclusions] The expression of HIF-1α and VEGF can be used to accurately assess the prognosis in ovarian cancer, and may provide new therapy target.
