[目的]研究血管生成抑制剂TNP-470对小鼠腹水瘤细胞腹腔种植的影响。[方法]80只615小鼠腹腔内接种腹水瘤细胞株后,随机分为5组,然后分别腹腔内注射生理盐水(第1组),DDP(第2组)及不同剂量的TNP-470(第3、4、5组),观察瘤细胞株对各组小鼠的腹水生成,瘤细胞数及生存时间;透射电镜观察TNP-470作用后腹腔内肿瘤细胞的形态学变化。[结果]与生理盐水对照组相比,DDP、不同剂量的TNP-470可显著抑制615小鼠腹水的生成,减少瘤细胞数量,延长生存期, DDP组、TNP-470低剂量组、TNP-470中剂量组、TNP-470高剂量组生存时间分别为26.25、19.75、28.38、21.25 d,与对照组生存时间14.88 d相比,有显著性差异(P<0.05),且中剂量TNP-470可使小鼠寿命明显延长,平均可达28.38 d。[结论]TNP-470能抑制小鼠腹水瘤腹腔种植及腹水生成,不同程度的延长生存期。 |

Abstract: |
[Objective]The aim of this study to observe the effect of TNP-470 on mice implanted intraperitoneally with ascitogenous tumor cells and investigate its mechanism. [Methods]Eighty 615 mice were injected with ascitogenous tumor cell line and were divided into 5 groups at random: Group 1,normal saline; Group2 , Cisplatin; Group 3~5, various concentration of TNP-470. Production of ascites, number of tumor cells and survival period were evaluated. Morphological changes of tumor cell were observed with eletromicroscope. [Results]Cisplatin and various concertration of TNP-470 obviously inhibited the production of ascites and prolonged the survival period of tumor-loading mice. Survival period of cisplatin,low dose TNP-470,middle dose TNP-470 and high dose TNP-470 was 26.25,19.75,28.38,21.25 d respectively. Comparison of above groups with group 1 showed significantly difference(P<0.05).The middle dose of TNP-470 obviously prolonged the survival period of tumor-loading mice, the survival period was 28.38 d. [Conclusions]TNP-470 could inhibit or eradicate mice following intraperitoneal implatation of ascitogenous tumor cells and production of ascites and prolong the survival period of mice that suffered from malignant ascites. |
