多西他赛和顺铂对大鼠脊髓损伤的形态学观察

摘要:
[目的]探讨化疗药物多西他赛(docetaxel)和顺铂对大鼠脊髓损伤的影响。[方法]将24只Wistar品系大鼠随机分为空白组、多西他赛组、顺铂组、顺铂和多西他赛合用组,腹腔注射单次给予多西他赛5 mg/kg、顺铂5 mg/kg、多西他赛和顺铂各5 mg/kg。5个月后,行活体灌注固定,取材,光镜和电镜观察脊髓的形态学变化。[结果]光镜下观察与对照组相比,顺铂组:灰质神经元形态未见异常,但可见血管扩张明显,白质神经纤维髓鞘变性肿胀断裂;多西他赛组:灰质神经元肿胀,尼氏体减少并消失,神经纤维变性坏死,神经胶质细胞增生,白质神经纤维髓鞘变性肿胀断裂;顺铂与多西他赛合用组:灰质内胶质细胞核固缩,灰质神经纤维变性肿胀,空泡样变,血管扩张,神经元尼氏体明显,白质神经纤维髓鞘变性肿胀断裂。电镜下,与对照组相比,顺铂组部分髓鞘的板层结构松解、扭曲;多西他赛组部分髓鞘结构出现板层结构松解、扭曲等轻度变化,细胞器相对完好;多西他赛与顺铂联合组髓鞘结构出现板层结构松解、扭曲。[结论]脊髓损伤后神经细胞损伤在形态改变上呈现多样性。化疗药物多西他赛和顺铂明显加重脊髓损伤,且顺铂与多西他赛合用脊髓损伤更重。

Abstract:
[Objective]To explore the injury of normal spinal cord induced by the docetaxel and cisplatin. [Methods]In this experiment, 40 Wistar rats were divided into following groups: normal, Docetaxel, cisplatin, combination of Docetaxel and cisplatin. Docetaxel was administrated singlely 5mg/kg. cisplatin was administrated singlely 5 mg/kg, docetaxel and cisplatin were administrated singlely 5mg/kg . Rats from each group were sacrificed at 5 months after the administrated by docetaxel and cisplatin were administrated singlely 5 mg/kg. The Paraflin sections of spinal cord were examined by microscope, and superthin sections were examined by transmission electron microscope. [Results]Under the light microscope, compared to normal group, some of the myelinated nerve fibers were degenerated and swollen in white matter with expansive blood vessels and neurons of no any change in gray matter in cisplatin group; swollen neuron and decreased nissl body in number and disappear, nerve fibers were degenerated and necrosised, neuroglia cells increase in number, myelinated nerve fibers were degeneration and swollen in white matter in docetaxel group, nuclear of neuroglia cell becomes small, myelinated nerve fibers were degeneratd and swollen in gray matter with vesicle degeneration , blood vessels expand , nissil body of neuron is obvious , nerve fibers were degeneration and swollen in white matter in the group of combination of Docetaxel and cisplatin, and under electron microscope , compare to normal group , in cisplatin group ,the lamina of the myelin sheaths was incompact and twisting; in docetaxel group , the lamina of the myelin sheaths was little incompact and twisting partly , the structure of organnels is intact, in the group of combination of Docetaxel and cisplatin, the lamina of the myelin sheaths was incompact and twisting .[Conclusions]The combination of docetaxel and cisplatin deteriorated the spinal cord injury of rats, especially in cisplatin plus docetaxel group.
