[目的]研究糖尿病大鼠不同病程心肌病变使心肌细胞内肌动蛋白含量的变化和超微结构的改变。[方法]①制造糖尿病大鼠心肌模型随机分组。②心肌免疫组织化学染色。③采用显微图像分析仪进行积分光密度分析,计算心肌型肌动蛋白平均积分光密度。④心肌病理改变的透射电镜观察。[结果]糖尿病病程6个月组心肌细胞心肌型 α-actin蛋白表达减少明显高于3个月以内组(4.06±2.03)病程3个月之后α-actin蛋白表达较健康鼠明显减少(145.8±30.66)。糖尿病大鼠心肌细胞心肌型α-actin蛋白分布不均,着色呈浅黄色,心肌纵切面可见α-actin蛋白表达主要分布于心肌肌膜处。心肌细胞核邹缩,线粒体肿胀,模糊,闰盘不连续,α-actin蛋白表达明显减少,有糖原沉积现象。[结论]糖尿病α-actin KNA减少改变了心脏能力的基础代谢和基因表达。 |

[Objective] To investigate pathologic changes of actin contents in myocardial cell and changes of supper micro-structure. [Method] ①Cardiac muscle model of DM rats divided groups at random.②Myocardial immunehistochemical stain.③Adopt micrograph analytical instrument to make a IOD analysis figure out cardiac-type α-actin IOZ.④Specimen for trans-mission electron microscope preparation. [Result]Cardiac-type α-actin expression of experimental rats decrease obviously after six months group had much more than those of three months(4.06±2.03).The cardiac-type α-actin expression subside obviously than healthy rats after three months(145.8±30.66). The cardiac-type α-actin expression subside obviously than healthy rats after six months(149.86±32.69). Cardiac-type α-actin of DM rats have shown out in order.The myocardial mitochondria have already shown not distinct,the intercalated disk distorted,cracked and big gap, the cell nucleus are malformation,All in all, cardiac-type α-actin expression decrease along with the disease headway gradually.The cell nucleus and endoplasm have large quantity of glucogenic accumulating phenomenon. [Conclusion] DM α-actin KNA decline changed basal metabolism and gene expression of cardiac energy. |
