摘要: |
[目的]研究顺铂(DDP)与5-氟尿嘧啶(5-FU)在合并用药前后对体外培养的人胃癌细胞的影响。[方法]采用MTT法观察体外培养的人胃癌细胞系SGC-7901在DDP与5-FU合并用药前后细胞存活率的差异,并通过流式细胞仪检测细胞周期变化, 透射电镜观察细胞超微结构变化。[结果] DDP与5-FU合用与单独应用化疗药相比细胞存活率明显下降,其增效倍数为单独用药的2.73~4.48倍(P<0.01), 细胞生长停止在S期,S期数分数为0.51,有效抑制了细胞有丝分裂及DNA合成, 细胞超微结构显示:联合用药后,细胞核固缩、空泡形成、微绒毛减少、线粒体及内质网肿胀。 [结论]结果表明顺铂与小剂量化疗药合用,可获得强于化疗药单独用药的疗效,5-氟尿嘧啶增加了化疗药物对肿瘤细胞的毒性,增加了细胞对药物的敏感性,降低了药物的副作用。 |

[Objective] To study the influences of the application of the cisplatin combined with 5-fluorouracil on human gastric carcinoma cell lines in vitro. [Methods]MTT method was used to investigate the cytotoxicity of the drugsagain human gastric carcinoma cell line SGC7901 in vitro.individually or in combinations. Light and electron microscopes were employed to observe the changes in morphology of the cells treated with the drugs. Flowing cytometry was applied to study the effects of the drugs on the cell cycle of the cell lines. [Results] DDP significantly decreased the survival level of cells with 5-FU compared with that the two drugs used individually, the potentiation of the combinations was ranged from 2.73 to 4.48 (P<0.01). The growth of cells stopped at Period S. The mitosis and the formation of DNA were restrained. The super-microstructure of cells showed that nucleonic shrinkage and necrosis ,swelling of mitochondria, dilation of endoplasmic reticulum and some other evidence appeared in the treated cells. [Conclusion] 5-FU can increase the sensitivity of carcinoma cells to kinds of DDP, and so reduce their side effects. |
