生长抑素对肝癌大鼠血管内皮生长因子的影响及病理学改变

摘要:
[目的]探讨生长抑素对大鼠肝癌细胞血管内皮生长因子产生能力及生长的影响。[方法] 取腹腔接种肝癌瘤株( CBRH-3 )大鼠 20 只,随机分为实验组和对照组,实验组每只大鼠在接种肿瘤细胞 24 h 后,腹腔注射人工合成生长抑素类似物 - 奥曲肽( 100 μg · kg-1 · d-1),连续 7 d ;对照组大鼠,腹腔注射生理盐水 0.5 mL/ d, 连续 7 d 。 第 8 天处死大鼠,测量肿瘤大小,免疫组织化学法检测肿瘤血管内皮生长因子( VEGF )的表达情况。[结果]肿瘤平均体积实验组为 1.26 cm3 明显小于对照组 2.57 cm3 ( P <0.05 );与对照组相比实验组肿瘤细胞坏死明显;实验组的 VEGF 表达积分 5 分明显低于对照组的 9 分( P <0.05 )。[结论] 生长抑素对大鼠肝癌的生长具有抑制作用,这种抑制作用可能通过抑制血管生成完成。

Abstract:
[Objective] To study the inhibitory effect of somatostatin to growth and production of vascular endothelial growth factor(VEGF) of hepatocellularcarcinoma (HCC). [Method] Twenty rats injected with hepatocellular carcinoma cells,CBRH-3,were randomly divided into treat group and control group. The animals in treat group were administered with octreotide(100 μg·kg -1· day-1 ) for 7 days, and those in control group with saline(0.5 mL/day).Till 8th day, the animals were terminated. The tumor volume and morphology of every case were recorded and the protein expression of VEGF was detected by immunohistochemistry. [Result] The mean tumor volume of treat group being 1.26 cm3was significantly smaller than that of control group being 2.57 cm3 ( P <0.05) .Under microscope, the more necrosis of carcinoma cells were observed in treat group than those in control. The expression point of treat group were 5 points being lower than that of control being 9 (P <0.05). [Conclusion] Octreotide has inhibition to the growth of rat HCC .This effect may be completed by inhibiting vasculogenesis.
