[目的] 本研究通过观察大鼠卵巢子宫组织中的雌孕激素受体水平,探讨米非司酮对妊娠和非妊娠状态下子宫卵巢功能影响的作用机制。[方法]性成熟而未受孕的雌性 Wistar大鼠共 30 只,随机分为对照组、药流组、未孕组,每组各 10 只。对照组给予精制色拉油;药流组于受孕后第 7 天分别给予米非司酮 8.3 mg/kg 和米索前列醇 100 μg/kg 灌胃;未孕组于动情前期给药,方法及剂量同药流组。所有大鼠均于给药后的第 3 个动情周期的动情前期处死,取出卵巢、子宫组织,用免疫组织化学染色方法检测雌孕激素受体水平。[结果] 对照组子宫和卵巢的孕激素受体阳性积分分别为 26 、23 ,药流组为 8 、12 ,未孕组为 17 、15 ;对照组子宫和卵巢的雌激素受体阳性积分分别为 26 、26 ,药流组为 16 、15 ,未孕组为 10 、11 。未孕组和药流组子宫及卵巢组织的雌孕激素受体阳性积分与对照组相比均显著减少( P<0.05);药流组子宫内膜的孕激素受体表达显著低于未孕组( P<0.05 )而药流组的子宫内膜雌激素受体和卵巢的雌孕激素受体与未孕组无显著性差异( P>0.05 )。[结论] 米非司酮对卵巢和子宫组织的雌孕激素受体均具有向下调节的作用;米非司酮对妊娠状态的子宫内膜孕激素受体的影响更为显著;上述影响在给予米非司酮两个动情周期之后,依然没有恢复正常。 |

Effect of mifepristone to the estrogen receptor (ER) and pregesterone receptor (PR) of ovary and uterus in rats |

[Objective] To investigate the mechanism of mifepnistone by studying the effect of mifepenistone(RU486) to the pregesterone receptor(PR) and estrogen receptor(ER) level of ovary and uterus in unpragnant and pragnant rats. [Methods] Thirty female Wistar rats were divided into 3 groups randomly, 10 rats in each group: 1. control group; 2. medication abortion group; 3. unpragnant group. Refined salad oil was administrated in group 1. RU486(8.3 mg/kg) and misoprostol(100μg/kg) were administrated by introgastric administration in seventh day after therat conceived in group 2. With the same medicine and measure as that in group 2,rats in group 3 were administrsted in the period of pro-oestrus. All rats were put to death in the prophase of the third estrus cycle after being administered,the ovaries and uterus were extracted out and analyzed by immunohistochemistrymethod. [Results] The positive accumulated point of PR of uterus or ovatry is 26 、 23 in group 1, 8 、 12 in group 2, 17 、 15 in group 3. The point of ER is 26 、 26 in group 1, 16 、 15 in group 2, 10 、 11 in group 3. The positive accumulated point of PR or ER of ovatry or uterus in group 2 or 3 was lower significantly than that in group 1 (P<0.05). The point of PR of uters in group 2 was lower significantly than in group 3 (P<0.05). The point of ER of uters or PR of ovary and uterus had no significant difference between group 2 and 3. [Conclusion] Ru486 havemuch more effect to PR of pragnant endometrium. Ru486 had down-regulation effectto ER or PR of ovary. |
