“心肌样”微环境中骨髓基质细胞分化为心肌样细胞

摘要:
[目的]探讨“心肌样”微环境中,骨髓基质细胞(marrow stromal cells,MSCs)是否可以分化为心肌样细胞。[方法]将SD成年大鼠的MSCs分离、培养后用5-BrdU标记,再将标记的MSCs与SD大鼠乳鼠心肌细胞(cardiomyocytes,CM)共培养,在倒置显微镜下观察MSC s形态学变化;用免疫细胞化学法检测MSCs中心肌特异性肌钙蛋白T。[结果]MSCs 3d后贴壁生长,呈梭形或多角形,细胞核大,细胞增殖迅速,当细胞80%~90%融合时加入CM, 共培养3d, MSCs变成长杆状,即呈现较为一致的极性排列。取共培养3周的细胞进行免疫细胞化学染色,见到经5-BrdU标记的部分MSCs在荧光显微镜下细胞核显现红色荧光的同时光镜下胞浆中心肌特异性肌钙蛋白T呈阳性。[结论]在“心肌样”微环境中,确实有部分MSCs转化为心肌样细胞。

Abstract:
[Objective]To investigate if marrow stromal cel ls(MSCs) could differentiate to cardiac-like cells in a “cardiac-like” milieu. [Methods] MSCs were s eparated, cultivated, and labeled with 5-BrdU, then cocultured with cardiomyocyt es(CM).The MSCs morphology changes were observed by invert microscope and the my ocardium specific troponinT were detected by immunocytochemistry. [Results]MSCs were adherent to dishes after 3 days of incubation with a shape of spindle or multiangle and a thophic nucleas, which proliferated rapidly with a 80%~90% co nfluence. In this moment, cocultured with CM, MSCs had a tendency to align with one another in close approximation. The MSCs labeled with 5-BrdU had red nuclei under fluorescence microscope and brown-pigmented cytoplasm under light microscope. [Conclusion] MSCs may differentiate to cardiac-like cells under “card iac-like” milieu in vitro. In the differentiation, the “cardiac-like” milieu may play a crutial role.
