[目的] 研究重组人干扰素α-2b(recombined human Interferon α-2b, rhIFNα-2b)含片对小鼠免疫功能的影响。[方法] 应用比色法、计数法及ELISA双抗体夹心法,分别测定在高(50 000 IU/只)、中(5 000 IU/只)、 低(500 IU/只)3种剂量rhIFNα-2b含片作用下,不同时间(10、20、30 d)小鼠脾脏NK细 胞毒指数、腹腔巨噬细胞吞噬百分率及吞噬指数和唾液IgA 含量的变化。[结果] 20 d各 剂量组NK细胞毒指数分别为(75.38±11.25)、(80.75±9.86)、(77.23±13.10)%,高于 空白基质片组(46.95±10.42)% (P<0.001); 20 d各剂量组吞噬指数分别为(0.619 ±0.054)、(0.664±0.046)、(0.599±0.038)%,高于空白基质片组(0.396±0.021)%(P<0.001);20 d 各剂量组唾液IgA含量分别为(70.88±10.38)、(66.28±8.98)、(54.4 1±6.35)μg/mL ,高于空白基质片组(35.67±3.51)μg/mL (P<0.001)。[结论] rhIFNα-2b含片具有提高小鼠免疫功能的作用。 |

[Objective] To study the effect of the contain of recombinant human interferon α-2b (rhIFNα-2b)on the immune function of mouse. [Methods] Cytotoxic index of the NK cells, phagocytic percentage and index of the abdomen macropha ges and IgA level in saliva in mouse treated with high-,moderate- or low-dosage of rhIFNα-2b(50 000,5 000,500 IU per mouse) were determined at different times (10 d, 20 d, 30 d) by colorimetry, count and sandwich ELISA, respectively. [Results] Compared with the blank matrix group,the cytotoxic index of 20 days is ( 46.95±10.42)%, the cytotoxic index of the NK cells of the three dosage groups of 20 days are (75.38±11.25),(80.75±9.86),(77.23±13.10)% respectively(P<0.001). Compared with the blank matrix group (the phagocytic index of macrophages of 20 days is 0.396±0.021), the phagocytic index of macrophages of the three dosage g roups of 20 days are (0.619±0.054), (0.664±0.046), (0.599±0.038) respectively(P<0.001). Compared with the blank matrix group, the IgA level in saliva of 20 days is (35.67±3.51) μg/mL the IgA level in saliva of the three dosage groups of 20 days are (70.88±10.38),(66.28±8.98), (54.41±6.35) μg/mL res pectivel y(P<0.001). [Conclusion] The contain of rhIFNα-2b has a significant role on enhancing the immune function of mouse. |
