细胞因子激活的脐血单核细胞对K562细胞株杀伤作用的观察

摘要:
观察及评价白细胞介素2(IL-2)及干扰素-α(IFN-α)激活的脐血单核细胞对白血病细胞株K562 的杀伤作用。[方法] 将细胞因子激活后的各组单核细胞(Mφ)与K562细胞株按一定效靶比作用48 h,采用MTT法检测各组的细胞毒效应。[结果] 单用IL-2和单用IFN-α刺激的脐血单核细胞与K562细胞株作用(效靶比20∶1), 其杀伤率分别为(8.31±1.32)%、(8.75±0.85)%。IL-2联合IFN-α激活脐血单核细胞对细胞株的杀伤作用明显增强,效靶比20∶1与50∶1时,杀伤率为(41.52±2.33)%(52.66±3.50)%,与单用IL-2及单用IFN-α组比较,具有显著性差异,P<0.01,并提示杀伤率随效靶比的增加而增加。 [结论] IFN-α和IL-2两者联合激活后的脐血Mφ对白血病细胞株的杀伤作用明显增强,杀伤率呈效靶比依赖关系。采用激活的脐血Mφ可用于干细胞移植时清除自体骨髓和外周血内的微小残留病变

Abstract:
To evaluate the cytotoxicity of cord blood monocyte- macrophage(Mφ) activated by interleukin-2(IL-2) and interferon-α(INF-α) and IL-2 in conbination with INF-α against K562 leukemic cell line.[Methods] The cytotoxicity of Mφ was measured with MTT assay.[Results] Mφ activated by 1 000 u/mL IL-2 alone and 1 000 u/mL INF-α alone produced significant cytotoxic activities against K562 cell, which were (8.31±1.32)% and (8.75±0.85)%, respectively(ratio of effector/target was 20∶1). Moreover, the same dose of IL-2 in conbination with INF-a could enhance the significant cytotoxic activities of Mφ, which was (41.52±2.33)% against K562. When the ratio of effector/target was 50∶1,the cytotoxicity was (52.66±3.50)%. With the ratio of effector/target increasing ,the cytotoxic activities of Mφ were enhanced, P<0.01.[Conclusions] The antitumor activity of Mφ could be further increased when IL-2 was in conbination with INF-a. And with the ratio of effector/target increasing ,the cytotoxicity of Mφ are enhanced. These results indicate that activaty cord blood monocyte-macrophage could be applied to get rid of micro-remain disease of leukemia and other malignant hematologic diseases
