鼻窦低氧环境对鼻息肉内成纤维细胞分泌VEGF的作用研究

摘要:
目的 通过缺氧刺激鼻息肉内成纤维细胞来观察VEGF的表达。方法 选取慢性鼻窦炎伴鼻息肉患者16例。测量慢性鼻窦炎患者鼻窦内氧分压,并以患者鼻息肉为材料,提取成纤维细胞,并在低氧环境刺激下培养。酶联免疫吸附法(ELISA)检测血管内皮生长因子(VEGF)的表达。结果 慢性鼻窦炎伴鼻息肉患者鼻窦平均氧分压47.74(36.72,56.84)mmHg。6例存在上颌窦内积液患者的鼻腔分泌物中VEGF蛋白的含量明显高于鼻腔分泌物和血浆(P<0.05,P<0.005)。息肉内提取的纤维原细胞在低氧环境下培养10、20 h产生的VEGF比普通氧分压环境下明显增多,差异有统计学意义(P<0.05)。结论 慢性鼻窦炎患者鼻窦内存在低氧的病理环境,低氧促进了鼻息肉成纤维细胞中VEGF的产生。

Effect of hypoxia environment in the sinuses on VEGF secretion from nasal polyp fibroblast cells

Abstract:
Objective In this study, we evaluated the VEGF expression in nasal polyp fibroblast cells by stimulation of hypoxia. Methods Measure the oxygen partial pressure in sinuses of 16 patients with chronic sinusitis, obtain fibroblast cells from nasal polyp of patients with chronic sinusitis and cultured them in a hypoxic environment. The expression of vascular endothelial growth factor (VEGF) was assessed by enzyme-linked immunosorbent assay (ELISA). Results The mean partial pressure of oxygen was 47.74 (36.72, 56.84) mmHg in patients with chronic sinusitis and nasal polyps. The VEGF protein content in nasal secretion of 6 patients with maxillary sinus effusion was significantly higher than that in nasal secretion and plasma (P<0.05, P<0.005). Compared with the normal oxygen partial pressure environment, the VEGF production of fibroblasts extracted from polyps in hypoxia environment for 10 h and 20 h was significantly increased (P<0.05). Conclusion There is a pathological environment of hypoxia in the sinuses of patients with chronic sinusitis.Hypoxia enhanced the production of VEGF in nasal polyp fibroblast cells.
