乳腺癌中Wnt信号通路成分NKD1、β-catenin和Cyclin D1蛋白的表达及意义

摘要:
目的 探讨NKD1、β-catenin和Cyclin D1在乳腺癌中的表达及意义。方法 选取2010年至2016年大连市妇女儿童医疗中心和大连市友谊医院病理科30例正常乳腺组织和124例乳腺浸润性癌非特殊型标本及临床资料。采用免疫组化法检测标本中NKD1、β-catenin和Cyclin D1蛋白的表达,并分析其与临床病理参数之间的关系。结果 在乳腺癌组织中NKD1阳性表达率30.65%;β-catenin异常表达率72.58%; Cyclin D1阳性表达率为54.03%,与正常乳腺组织比较,差异均有显著性意义(P均<0.05)。NKD1和β-catenin表达与乳腺癌组织学分级、pTNM分期及淋巴结转移有关,Cyclin D1表达与乳腺癌pTNM分期及淋巴结转移亦有关,且NKD1表达缺失还与ER状态相关,差异均有统计学意义(P均<0.05)。NKD1与β-catenin和Cyclin D1表达呈负相关(r分别为-0.454,-0.192,P均<0.05)。结论 NKD1、β-catenin和Cyclin D1蛋白表达与乳腺癌侵袭和转移密切相关,联合检测3种蛋白有助于判断乳腺癌生物学行为和评估预后。

Abstract:
Objective   To investigate the clinical significance of NKDl,β-catenin,Cyclin D1 protein expression in Breast carcinoma.  Methods   Thirty normal breast tissues and 124 non-specific invasive breast cancer tissues were collected from Dalian Municipal Woman and Children’s Medical Center and Dalian Friendship Hospital. Immunohistochemistry was used to detect the expressions of NKDl,β-catenin,Cyclin D1 protein. Spss17.0 software was used for statistical analysis. Results   In breast cancer tissues, the negative frequency of NKD1 expression was 69.35%, the abnormal expression frequency of β-catenin was 72.58%, and the positive frequency of Cyclin D1 expression was 54.03%. There were significant differences between normal breast tissures and breast cancer tissues. Down-regulation of NKD1 expression and abnormal expression of β-catenin were significantly correlated with histologic grades, pTNM stages and the lymph node metastasis of breast cancer (P<0.05). Up-regulation of Cyclin D1 was significantly correlated with pTNM stages and the lymph node metastasis of breast cancer (P<0.05). NKDl expression was negatively correlated with β-catenin abnormal expression and Cyclin D1 expression (r=-0.454,r=-0.192,P<0.05).  Conclusions   NKD1,β-catenin,Cyclin D1 protein could become indicators of prognosis in breast cancer.
