目的 研究氨肽酶N(aminopeptidase N,APN)在乳腺癌中的表达及临床意义,并观察其与基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)、基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)的关系。方法 收集河北省人民医院病理科2011年7月至2012年9月乳腺癌石蜡标本60例,癌旁正常乳腺组织石蜡标本20例,及患者的临床病理资料。采用免疫组织化学SP法检测APN在80例标本中的表达,并分析APN表达与乳腺癌患者临床病理特征之间的关系。检测MMP-2、MMP-9在乳腺癌组织中的表达,及与APN表达的相关性。结果 APN在乳腺癌中的阳性表达率36.7%(22/60)明显高于癌旁正常乳腺组织0%(0/20)(χ2=10.115,P=0.001)。APN在乳腺癌中的表达与肿瘤病理类型有关(P=0.035),多表达于浸润性导管癌;与患者绝经状况、肿瘤大小、临床分期、组织学分级、淋巴结转移状态、ER、PR、Her-2等无关(P>0.05)。乳腺癌组织APN的表达与MMP-2 、MMP-9的表达无相关性(P>0.05)。结论 APN在乳腺癌中的表达高于正常乳腺组织,但APN在乳腺癌中侵袭转移的机制有待进一步深入研究。 |

Objective To investigate the significance of Aminopeptidase N (APN) expression in breast cancer, and to discuss the role of APN in the process of breast carcinoma invasion and metastasis by analyzing the relationships between APN and Matrix metalloproteinase-2 (MMP-2), Matrix metalloproteinase-9 (MMP-9). Methods Immunohistochemistry (SP method) was performed to detect the expressions of APN, MMP-2 and MMP-9 in 60 cases of breast cancer tissues, as well as APN expression in 20 cases of adjacent normal breast tissues. Results The positive frequency of APN expression in breast cancer tissues (36.7%, 22/60) was significantly higher than that in adjacent normal breast tissues (0%, 0/20) (χ2=10.115, P=0.001). APN expression was associated with tumor pathological types (P=0.035). However, there was no relationship between the APN expression and other clinicopathological features, such as menopausal state, clinical stage, tumor size, histological grade, nodal status, ER, PR, and Her-2 (P>0.05). There was no significant relationship between the APN expression and MMP-2, MMP-9 expression (P>0.05). Conclusion APN expression was higher in breast cancer than that in normal breast tissues, but it needs further study if APN expression could play a role in invasion and metastasis in breast cancer. |
