大鼠脊髓损伤程度与血清β-淀粉样前体蛋白及超敏C反应蛋白的相关性研究

摘要:
[摘要] 目的 探讨大鼠急性脊髓损伤(SCI)程度与血清β-淀粉样前体蛋白(β-APP)和超敏C反应蛋白(hs-CRP)含量变化的相关性。 方法 取雌性SD大鼠90只,采用动脉夹压力法制备大鼠脊髓轻度损伤(轻度损伤组,30只)和重度损伤(重度损伤组,30只)动物模型,另30只大鼠仅行椎板切除作为假手术组。通过行为学观察(BBB评分)了解脊髓损伤程度及变化,分别于造模成功后12 h、24 h、3 d 、7 d、14 d采股静脉血,ELISA检测不同程度SCI大鼠伤后血清β-APP、hs-CRP含量变化。 结果 重度脊髓损伤组与轻度损伤、假手术组各时间点BBB评分差异有显著性意义(P<0.05)。β-APP于损伤后24 h达到高峰后逐渐回落,hs-CRP于3 d达到高峰后回落。重度脊髓损伤组与轻度脊髓损伤、假手术组各时间点β-APP、hs-CRP比较差异均有显著性意义(P<0.05);轻度脊髓损伤组与假手术组β-APP、hs-CRP水平除14 d点差异无显著性意义外,其余各时间点差异均有显著性意义(P<0.05)。 结论 急性SCI大鼠血清β-APP、hs-CRP水平变化可反映SCI的严重程度,β-APP比hs-CRP的变化敏感。

Relation between the degree of the acute spinal cord injury and the serum β- amyloid precursor protein and hypersensitive-cross-reactive protein levels

Abstract:
[Abstract] Objective To investigate the relation to the degree of the acute spinal cord injury (SCI) by detecting the serum β-amyloid precursor protein (β-APP) and hypersensitive-cross-reactive protein (hs-CRP) levels after acute spinal cord injury in rate at different time points. Methods Ninety healthy adult female SD rats were randomly assigned into 3 groups. The bulldog clamp was used to create models of severe SCI (group A) and mild SCI (group B). In group C, only laminectomy was performed without SCI. The ethological study (BBB scores) was used to learn the degree of the acute spinal cord injury. The serum was collected at 12 h, 24 h, 3 d, 7 d, and 14 d after operation through femoral vein. Serum β-APP and hs-CRP were detected by ELISA. Results The serum β-APP and hs-CRP reached the peak at 24 h and 3 d, respectively after acute SCI. Serum β-APP and hs-CRP levels in SCI groups (A and B) were both significantly higher than those in without SCI group (C) at all time points (P<0.05), except at 14 d between group B and group C. Conclusions The serum β-APP and hs-CRP levels are positively associated with the severity of SCI. β-APP would be a more sensitive indicator than hs-CRP in a certain time. A acute SCI model of SD rats can be established by bulldog clamp, in order to mimic the clinical SCI.
