骨形态发生蛋白4对反应性星形胶质细胞增殖的影响

摘要:
[摘要] 目的 利用成年SD大鼠脊髓损伤原代培养的反应性星形胶质细胞模型,探讨骨形态发生蛋白4(BMP4)与反应性星形胶质细胞增殖之间的关系。 方法 建立成年SD大鼠脊髓损伤原代培养的反应性星形胶质细胞模型,用BMP4(10 ng/mL)和BMP4的拮抗剂——Noggin(100 ng/mL)处置反应性星形胶质细胞48 h,通过免疫荧光的方法检测各实验组(正常对照组、BMP4组、Noggin组及BMP4+Noggin组)中星形胶质细胞的标记分子vimentin Brdu,观察BMP4对反应性星形胶质细胞增殖的影响。 结果 BMP4组中Brdu阳性细胞占星形胶质细胞的平均百分比为14.03 %,明显高于正常对照组(3.10%),P<0.01;而BMP4+Noggin组中Brdu阳性细胞数占星形胶质细胞的平均百分比为3.81%,明显低于BMP4组(P<0.01)。 结论 在成年SD大鼠脊髓损伤原代培养的反应性星形胶质细胞模型中,BMP4可刺激反应性星形胶质细胞的增殖。

Effects of BMP4 on the proliferation of reactive astrocytes

Abstract:
[Abstract] Objective To investigate the effect of BMP4 on proliferation of reactive astrocytes derived from the injured spinal cord of adult rat. Methods The reactive astrocytes from the injured spinal cord were isolated and purified, then treated respectively with 10 ng/mL BMP4, 100 ng/mL Noggin (BMP4 antagonist) and BMP4 combined with Noggin for 48 h. Immunofluorescence (IF) was used to detect the expression of vimentin (astrocyte marker) and Brdu in reactive astrocytes of different groups (Control group, BMP4 group, Noggin group and BMP4+Noggin group). Results The mean percentages of Brdu+ astrocytes (vimentin+) in BMP4 group (14.03%) were higher than that in normal group (3.10 %, P<0.01) and that in BMP4 + Noggin group (3.81%, P<0.01). Conclusion  BMP4 may promote the proliferation of reactive astrocytes from the injured spinal cord.
