[摘要] 目的探讨Livin基因在结直肠癌中的表达及其与PTEN和Caspase-3的相关性。方法 收集结直肠癌、腺瘤及大肠正常黏膜组织各50例,应用免疫组织化学SP染色方法检测Livin、PTEN及Caspase-3的表达情况并进行评分。结果 结直肠癌、腺瘤及大肠正常黏膜组织中Livin蛋白的阳性表达率分别为64.00%、56.00%、18.00%,结直肠癌组织中Livin蛋白的阳性表达率高于腺瘤及大肠正常黏膜组织;Livin蛋白阳性表达率与淋巴结转移及Duke分期有关,而与结直肠癌患者的年龄、性别、分化程度无关。在结直肠癌、腺瘤及大肠正常黏膜组织中PTEN蛋白的阳性表达率分别为40.00%、76.00%、98.00%,Caspase-3蛋白的阳性表达率分别为46.00%、84.00%、92.00%,结直肠癌组织中PTEN、Caspase-3蛋白的阳性表达率明显低于腺瘤及正常黏膜组织,差异有显著性意义(P<0.01);PTEN、Caspase-3阳性表达率与结直肠癌Duke分期及淋巴结转移密切相关,与结直肠癌患者的年龄、性别及肿瘤分化程度无关。在结直肠癌组织中Livin蛋白阳性表达与PTEN及Caspase-3蛋白表达均呈负相关。结论 凋亡抑制因子Livin、凋亡因子Caspase-3及抑癌基因PTEN参与了结直肠癌的发生、发展。在结直肠癌中PTEN蛋白活性被抑制加速了Livin促进结直肠癌的发生。Livin促进结直肠癌的发生可能通过抑制Caspase-3的活性实现。 |

[Abstract] Objective To explore Livin,PTEN and Caspase-3 protein expression and their correlationship in colon cancer. Methods The expressions of Livin,PTEN and Caspase-3 protein in 50 cases of colon cancer, colon adenoma and normal colorectal mucosa were detectded by immunohistochemistry staining. Results (1)The expression of Livin in colon cancer, colon adenoma and normal colorectal mucosa was 64.00%,56.00%,18.00%.Livin protein positive expression rates in colon cancer tissues was more than those in colon adenoma and nomal colorectal mucosa and had statistical significance. (2) The expression of PTEN in colon cancer, colon adenoma and normal colorectal mucosa was 40.00%,76.00%,98.00%. The expression of Caspase-3 in colon cancer, colon adenoma and normal colorectal mucosa was 46.00%, 84.00%, 92.00%. Caspase-3 and PTEN protein positive expression rates in colon cancer tissues were less than those in colon adenoma and nomal colorectal mucosa(P<0.01).(3) Caspase-3,PTEN protein expression had prominent inverse correlation with the Livin expression. Conclusion Livin,PTEN,Caspase-3 participate the happen and the development of colon cancer. The suppressed of PTEN protein may accelerate the express of Livin,and promote the happening of colon cancer. Livin promotes colon cancer through inhibited caspase-3 protein activity. |
