黄芪甲苷对两肾一夹型肾性高血压大鼠肾脏病变的影响

摘要:
目的 探讨黄芪甲苷对肾性高血压大鼠肾脏的影响。方法 将70只SD雄性大鼠分为2K1C组(n=60)与假手术组(n=10),2K1C组按照经典两肾一夹(2K1C)型肾性高血压大鼠模型方法建模,建模成功后随机分为模型组、低剂量黄芪甲苷组、高剂量黄芪甲苷组,每组20只。低剂量黄芪甲苷组和高剂量黄芪甲苷组分别给予黄芪甲苷200 mg/(kg·d)、800 mg/(kg·d),模型组和假手术组给予生理盐水200 mg/(kg·d)替代治疗,用药8周。处死大鼠,取右肾组织匀浆检测NO、NOS、肾素和AngII活性。手术前,各组大鼠尾动脉收缩压比较差异无显著性意义(P>0.05)。 结果 (1)术后8周,低剂量黄芪甲苷组、高剂量黄芪甲苷组大鼠尾动脉收缩压(134.2±8.2和128.3±7.8)mmHg明显低于模型组的(152.4±8.5)mmHg,但仍高于假手术组的(106.9±4.2)mmHg(P<0.05)。(2)光镜下可见模型组肾小球毛细血管塌陷,肾小球萎缩、硬化、坏死,部分肾小球代偿性肥大,肾间质可见大量炎症细胞和红细胞浸润等;高剂量黄芪甲苷组,可见肾小球肥大减轻,平均直径减小,未见肾小球萎缩、坏死,肾间质仍偶见炎症细胞浸润。 (3)模型组、低剂量黄芪甲苷组、高剂量黄芪甲苷组右肾组织匀浆NO、NOS明显低于假手术组(P<0.05);AngII含量显著高于假手术组(P<0.05)。低剂量黄芪甲苷组、高剂量黄芪甲苷组与模型组间上述指标比较差异有显著性意义(P<0.05)。 结论 黄芪甲苷能够有效降低肾性高血压大鼠肾脏损伤。

Effect of Astragaloside on renal lesions of 2K1C model of renovascular hypertensive rats

Abstract:
[Abstract] Objective To study the effect and mechanism of Astragaloside on renal lesions in 2K1C model of renovascular hypertensive rats. Methods Seventy male SD rats were randomly divided into 2K1C group (n=60) and sham operation group (n=10). Renovascular hypertensive model was built in 2K1C group according to 2K1C model method, and 2K1C group were further randomly divided into model group, low dose Astragaloside group, and high dose Astragaloside group. Low dose Astragaloside group and high dose Astragaloside group were given astragaloside 200 mg/ (kg·d) and 800 mg/ (kg·d), respectively. Model group and sham operation group were given normal saline 200 mg/ (kg·d). After 8 weeks' treatment, the rats were killed and NO, NOS and AngII renin activity of right kidney were measured. Before the operation, the rat tail artery systolic blood pressure showed no statistical difference (P>0.05).Results (1) Eight weeks after operation, low dose Astragaloside group, high dose Astragaloside group rat tail artery systolic blood pressure was significantly lower than that in the model group, but still higher than the sham operation group (P<0.05). (2) Histologically, model group showed glomerular capillary collapse, glomerular atrophy, sclerosis, necrosis, partial glomerular compensatory hypertrophy, interstitial inflammatory cell and red blood cell infiltration. In high dose Astragaloside group, glomerular hypertrophy alleviated, average diameter decreased, no glomerular atrophy or necrosis was seen, there was still occasional renal in terstitial inflammatory cell infiltration. (3) NO, NOS in Model group, low dose Astragaloside group, and high dose Astragaloside group measured in right kidney were significantly lower than those in sham operation group (P<0.05); whereas AngII was significantly higher than that in sham operation group (P<0.05). The above values were statistically different (P<0.05) among low dose Astragaloside group, high dose Astragaloside group and model group. Conclusion Astragaloside can effectively reduce the damage of kidney in renal hypertensive rats.
