梗阻型黄疸小鼠模型构建及相关指标观察

摘要:
目的 建立并动态观察小鼠梗阻型黄疸模型。方法 选取Balb/c小鼠40只,随机分为模型组(32只)和对照组(8只), 实验组结扎胆总管。于结扎后第1天,第3天,第5天和第7天动态观察胆总管直径、小鼠肝脏病理变化、血常规及肝功能变化。结果 实验结果表明,小鼠随着胆总管结扎时间的延长, 其胆总管的直径增宽,ALT,AST, TBIL,DBIL,ALP,CHOL,中间细胞总数百分数升高,均在术后第3天到达高峰,然后其数值有所下降。白细胞总数在术后第1天升高,第3天降到最低点,之后有所恢复至平稳,但是仍低于正常。淋巴细胞百分比术后下降,到第5天降到最低点。肝细胞坏死随术后时间长而加重。结论 Balb/c小鼠在通过结扎胆总管造成梗阻型黄疸时, 其变化的规律与人类相似,并且造模时间短,是研究梗阻型黄疸病理生理机制和治疗的可靠实验模型。

Abstract:
[Abstract] Objective To establish mouse biliary obstruction model and to study the model dynamically. Methods Forty Balb/c mice were used and divided into experimental group (n=32) and control group (n=8). Mice of the experimental group were inflicted with obstructive jaundice through ligation of common bile duct. Mice were sacrificed on the 1st, 3rd, 5th, 7th days after the ligation to measure bile duct diameter, complete blood count, liver function and to study morphological changes in the liver. Results The levels of common bile duct diameter, MID%, ALT, AST, TBIL, DBIL, ALP and CHOL elevated in association with the time of common bile duct ligation, reached the peak on the third day after surgery, then began to decline. WBC elevated on the first day after surgery, then began to decline. LYM% declined after surgery, to the minimum on the fifth day. Liver cell necrosis increased in association with surgery time. Conclusion The clinical manifestations of this model are similar to those of patients with biliary obstruction and the cycle time of inducing this model is short. This model might be valuable in researching the mechanism of the obstructive jaundice.
