非小细胞肺癌肿瘤细胞的E-cadherin和vimentin蛋白表达及病理意义

摘要:
目的 了解非小细胞肺癌(NSCLC)肿瘤细胞上皮-间质转化(EMT)情况,并探讨其病理意义。方法 用免疫组织化学方法检测143例NSCLC患者肿瘤细胞和44例患者癌旁支气管黏膜上皮细胞的上皮标志蛋白E-cadherin和间叶标志蛋白vimentin的表达情况,用SPSS 17.0 统计软件分析上述蛋白表达与患者部分临床病理参数关系。结果 E-cadherin阳性表达率在肺癌细胞为52.4%,显著低于癌旁支气管黏膜上皮细胞的70.5%(P<0.05),且癌细胞E-cadherin表达丢失与癌旁支气管黏膜上皮细胞表达丢失显著正相关(r=0.539,P<0.05);在低分化腺癌为34.4%,显著低于高、中分化腺癌的61.7% (P<0.05),在男性肺癌患者为43.2%,显著低于女性患者的64.5%(P<0.05),但其表达与患者的年龄、吸烟状况、淋巴结转移、组织学类型无显著相关关系(P>0.05)。Vimentin阳性表达率在肺癌细胞为36.4%,显著高于癌旁支气管上皮细胞的20.5%(P<0.05),且癌细胞vimentin表达与癌旁支气管黏膜上皮细胞表达显著正相关(r=0.814,P<0.05);在低分化腺癌为62.5%,显著高于高、中分化腺癌的30.0%(P<0.05),但其表达与患者的性别、年龄、吸烟状况、淋巴结转移、组织学类型无显著相关关系(P>0.05)。E-cadherin与vimentin蛋白表达无显著相关关系(r=0.108,P>0.05)。结论 EMT现象存在于NSCLC,且在低分化腺癌表现尤为明显。

Abstract:
Objective 〖WTBZ〗To understand the situation of epithelial-mesenchymal transitions (EMT) in non-small cell lung cancer (NSCLC) and investigate the pathological and clinical significance of it. 〖WTHZ〗Methods 〖WTBZ〗The immunohistochemistry was used to detect protein expressions of E-cadherin and vimentin in 143 samples of NSCLC and 44 samples of bronchus epithelial cells surrounding tumor. The SPSS17.0 statistic software was used to analyze the correlation between the protein expressions and the clinic-pathological agents of the patients with NSCLC. 〖WTHZ〗Results 〖WTBZ〗The positive expression rate of E-cadherin protein in the tumor cells of NSCLC was statistically lower than that in bronchus epithelial cells surrounding tumor (52.4% vs.70.5%,P<0.05), and there was significant correlation between tumor cells and their corresponding bronchus epithelial cells surrounding tumor (r=0.539, P<0.05). Furthermore, the rate in the poorly differentiated adenocarcinoma(34.4%)was statistically lower than that in the well-moderately differentiated adenocarcinoma(61.7%) (P<0.05), and the rate in the male patients(43.2%) was statistically lower than that in the female (64.5%)(P<0.05). However,the expression was not significantly related to patients’ age, smoking status, tumor histological type and lymph node metastasis(P>0.05). The positive expression rate of vimentin protein in the tumor cells of NSCLC was statistically higher than that in bronchus epithelial cells surrounding tumor (36.4% vs 20.5%,P<0.05), and there was significant correlation between tumor cells and their corresponding bronchus epithelial cells surrounding tumor(r=0.814,P<0.05). The rate in the poorly differentiated adenocarcinoma(62.5%) was statistically higher than that in the well-moderately differentiated adenocarcinoma(30.0%)(P<0.05). The expression was not significantly related to patients’ gender, age, smoking status, tumor histological type and lymph node metastasis(P>0.05).The expression of E-cadherin protein was not related to that of vimentin (r=0.108,P>0.05).〖WTHZ〗Conclusion 〖WTBZ〗EMT phenomenon occurs in NSCLC, panticalarly in the poorly differentiated adenocarcinoma.
