[目的] 探讨甲型H1N1流感病毒疫苗对小鼠CD4 +CD25 +Foxp3分子表达的影响。[方法] SPF级BALB/c 小鼠72只,雌雄各半,用甲型H1N1流感病毒灭活疫苗免疫小鼠,按疫苗注射剂量随机分为高(0.3 mL)、中(0.2 mL)、低(0.1 mL)3组及对照组,注射后4、8、12、19、26、34天处死动物,用FACS分析小鼠脾脏Foxp3、CD4 +CD25 +和CD4 +CD25 + Foxp3三种分子表达变化。[结果] 不同疫苗注射剂量对三种分子表达平均值影响不同。Foxp3和CD4 +CD25 +平均值,0.1 mL组明显低于对照组、而0.3 mL组明显高于对照组(P<0.01);CD4 +CD25 +Foxp3平均值,0.1 mL组明显高于对照组(P<0.01)。随着造模时间的延长,各时间点分子表达平均值也不相同:Foxp3各时间点无明显差别;CD4 +CD25 +第34天明显低于第4天(P<0.05);CD4 +CD25 +Foxp3第19、26天明显高于第4天(P<0.05)。[结论] 应用甲流疫苗免疫可使小鼠CD4 +CD25 +Foxp3 T细胞免疫功能增强。 |

[Objective] To study the effection of influenza A(H1N1)virus vaccine on expression of CD4+CD25+ Foxp3 of mice.[Methods] Total 72 Balb/c mice of experimental SPF were used and divided randomly into high(0.3 mL), middle(0.2 mL) and low(0.1 mL)dose and control groups according to vaccine dose. These mice were immuned by influenza A(H1N1) inactivated vaccine and terminated at 4、8、12、19、26、34 day following vaccine. The CD4+, CD25+, Foxp3 expression of spleen cells were analyzed with FACS. [Results] The doses of injected vaccine effected on expressions of the three proteins.The Foxp3 and CD4+CD25+ average values in group 0.1 mL were significantly lower but higher in group 0.3 mL than those of control group(P<0.01);the CD4+CD25+Foxp3 average value in group 0.1 mL was significantly higher than that of control group(P<0.01). The average values of the three proteins were different in different time points. There were no difference in Foxp3,but the average values of CD4+CD25+ in 34th day was lower than that in 4th day significantly(P<0.05), the average values of CD4+CD25+ Foxp3 in 19th and 26th day were higher than that of the 4th day significantly(P<0.05).[Conclusion] Influenza A(H1N1) virus vaccine could increase CD4+CD25+ Foxp3 T cell immune function. |
