摘要:
[目的] 了解Rad18敲除雄小鼠生殖细胞的变化。[方法] 用免疫组织化学方法检测2、6、12月龄Rad18-/-雄小鼠生精小管生殖细胞的特异性标记物TRA98的表达及分布,并与野生(WT)雄小鼠比较;用原位杂交技术检测Rad18在1周龄WT雄小鼠生精小管未分化精原细胞中的表达。[结果] 缺失TRA98标记的早期精原细胞的生精小管比例在12月龄Rad18-/-和WT雄小鼠中分别为39.50%和2.76%,差异有显著性意义(P<0.05);但Rad18-/-退化生精小管中保留正常的Sertoli细胞,其附睾中未见TRA98阳性的生殖细胞,且精子形态正常;1周龄WT小鼠睾丸未分化精原细胞中有Rad18蛋白表达。[结论] Rad18可能对精原干细胞有维持作用。

Abstract:
[Objective] To investigate the function of Rad18 in germ cells of male mice. [Methods] Use germ line cell-specific marker TRA98 to detect the germ cells in the seminiferous tubules of 2-,6- and 12-month-old Rad18-/- mice. Use in situ hybridization to confirm the expression of Rad18 in the spermatogonia of 1-week wild type (WT) mice. [Results] The percentage of abnormal seminiferous tubules without spermatogonia in 12-month-old Rad18-/- testes was 39.5%,more than 2.76% in WT testes, P<0.05. The degenerated tubules in Rad18-/- testes retained normal Sertoli cells. No TRA98 positive germ cells entered the Rad18-/- epididymis. The spermatozoa found in the 12-month-old Rad18-/- epididymis displayed normal morphology. Rad18 was highly expressed in the spermatogonia of 1-week-old WT mice. [Conclusion] Rad18 is involved in maintaining spermatogonial stem cells.
