[目的] 通过观察亚慢性铅暴露对小鼠附睾组织Y染色体基因Ddx3y表达的影响,为探讨铅的雄性生殖毒作用机制提供依据。[方法] 昆明种小鼠48只,按体重将小鼠随机分为对照组、0.5 g/L、1.0 g/L和1.5 g/L 醋酸铅染毒组。通过自然饮用含不同浓度醋酸铅的方式使小鼠染铅,连续染毒60 d后取附睾组织。HE染色观察组织病理学变化,Western blotting和免疫组化方法检测附睾组织Ddx3y蛋白的表达及组织分布。[结果] 与对照组比较,染铅组附睾管内精子数和分泌液均减少,尤其 1.5 g/L染铅组最为明显。Western blotting检测结果显示,染铅组的小鼠附睾组织中Ddx3y蛋白表达明显低于对照组,且有剂量-反应关系。免疫组化结果显示,随着染铅剂量的增加,附睾组织中抗Ddx3y蛋白免疫反应明显降低。[结论] 亚慢性铅暴露显著下调小鼠附睾组织Y染色体基因Ddx3y蛋白表达。 |

[Objective] To investigate effects of subchronic exposure to lead on Ddx3y protein expression of Y chromosome in the epididymis of mice and provide target evidence for exploring the mechanism of lead-induced male reproductive toxicity. [Methods] Forty-eight mice were divided into 4 groups of 12 each. The mice in group 1 received drinking water alone (control), while those in group 2, 3 and 4 received 0.5 g/L, 1.0 g/L and 1.5 g/L lead acetate respectively through drinking. On the 60th day after lead exposure, the mice were decapitated and the epididymis were taken. The slices of the fixed epididymis tissues were performed with HE staining. Western blotting and immunohistochemistry were used to detect Ddx3y protein expression and tissue distribution. [Results] The groups exposed to lead showed the decreased in the canal of epididymis of mice. Western blotting results showed that expression of Ddx3y protein significantly decreased in the epididymis of mice in the lead groups. Immunohistochemistry results showed that the immune response intensity of unti-Ddx3y protein significantly decreased in the lead groups. [Conclusion] The subchronic exposure to lead can down-regulated the expression of Ddx3y protein in the epididymis of mice. It indicated that lead inhibiting maturation of sperm may be associated with the down-regulated the expression of Ddx3y protein in the epididymis. |
