青光眼是眼科临床中常见的致盲性眼病之一。目前的治疗方法主要是通过药物或手术降低眼压,对视网膜神经节细胞及其轴突损伤的保护也越来越引起人们的重视。随着对相关卷曲螺旋形成蛋白激酶通路(Rho/ROCK)生物学功能的深入研究,目前发现抑制Rho/ROCK通路可以增加小梁网途径的房水引流,降低眼内压,增加眼部血液供应,促进神经节细胞的存活。本文就Rho/ROCK通路与青光眼关系的研究进展进行综述。 |

Abstract: |
Glaucoma is one of the world’s leading causes of irreversible blindness.The common treatment of glaucoma has been aimed at lowering elevated IOP, either via drugs or operation.However,neuroprotection of ganglion cells and their axons has evoked more and more attention.Accumulating experimental evidence suggests that Rho kinase and Rho GTPase inhibitors can increase aqueous drainage through the trabecular meshwork, leading to a decrease in intraocular pressure.In addition to their ocular hypotensive properties, inhibitors of both ROCK and Rho GTPase have been shown to enhance ocular blood flow, retinal ganglion cell survival and axon regeneration.These properties indicate that targeting the Rho GTPase/ROCK pathway with selective inhibitors represents a novel therapeutic approach aimed at lowering increased intraocular pressure in glaucoma patients. |
