[目的]观察CO中毒小鼠脑海马区血管内皮系统Ang-1、Ang-2和Tie-2表达的变化规律,探讨CO中毒对小鼠脑血管内皮系统的影响。 [方法]采用逆转录聚合酶链反应(RT-PCR)测定空气对照组和CO中毒组小鼠(各48只)在不同时间点脑海马区Ang-1、Ang-2及Tie-2的mRNA相对表达量。[结果]与对照组比较,CO中毒后脑血管内皮系统Ang-1、Ang-1和Tie-2 mRNA表达均呈双峰性增高,双峰分别在CO中毒后3 d(7.02±0.11、7.67±0.22和8.73±0.07,P<0.05)和7 d(6.60±0.36、7.99±0.06和7.69±0.05,P<0.05)。 CO组的Ang-2/Ang-1比值在6 h达到第一高峰(1.55±0.07,与对照组比较P<0.05),2 d到达低谷,随后比值缓慢上升于第7天达到第二个高峰(1.24±0.10,与对照组比较P<0.05),14 d恢复正常。[结论]CO中毒后,小鼠脑内血管内皮Ang/Tie-2系统mRNA水平表达呈双峰性增高,第二个高峰可能与迟发性脑病相关。 |

[Objective] To detect dynamic change of mRNA expressions of Ang-1, Ang-2 and Tie-2 to reveal the effect of carbon monoxide (CO) on endothelial system of brain. [Methods] RT-PCR was used to detect mRNA dynamic change of Ang-1, Ang-2 and Tie-2 in the brains of mice in CO poisoning group and control group at different time points. [Results] Comparing with control group, after CO poisoning, Ang-1, Ang-2 and Tie-2 mRNA levels showed two peaks, respectively in 3 day (7.02±0.11, 7.67±0.22 and 8.73±0.07, P<0.05) and 7 day (6.60±0.36, 7.99±0.06 and 7.69±0.05, P<0.05). The ratio of Ang-2 to Ang-1 in CO poisoning group reached the peak at 6 h (1.55±0.07, compared with control group P<0.05), decreased bottom at 2 day, raised slowly to peak at 7 day (1.24±0.10, compared with control group P<0.05), and then decreased to normal at 14 day. [Conclusions] The expressions of Ang-1, Ang-2 and Tie-2 mRNA in endothelial system of mouse brain after CO poisoning have two peaks and the second peak might be related to delayed encephalopathy. |
