结石形成中骨桥蛋白与CD44的作用研究

摘要:
[目的]探讨骨桥蛋白(OPN)与CD44在泌尿系结石形成中的作用。 [方法]采用0.5%乙二醇+0.5%氯化氨饮水法建立高草酸尿症大鼠模型。HE染色光镜下观察肾小管腔内的结石晶体;钙盐染色光镜下观察肾小管上皮细胞基底膜和间质处的钙盐沉积情况;免疫组化染色观察OPN和CD44表达水平和位置,分析不同时段(第3、5、7、14、21、28天)结晶体与OPN和CD44表达分布及量变关系。[结果]肾小管OPN与CD44的表达随时间增加、钙盐沉积增多而增强,以后表达减弱;在第14天内,OPN与CD44表达保持正相关关系(P<0.05)。[结论]OPN和CD44表达与高草酸尿液状态的结石形成关系密切,可能成为Randall斑的起源。

Abstract:
[Objective] To investigate the mechanisms of osteopontin and CD44 in urinary stone formation of rat with hyperoxaluria. [Methods] The hyperoxaluria rat model was established by administration of 0.5% ethylene glycol and 0.5% ammonium chloride in drinking water for 3 to 28 day. Calculus crystals deposited in renal tubules were observed under light microscopy (HE-staining). Calcium depositon in the basement membrane of renal tubular epithelial cells and stroma was observed by calcium-staining. The expressions and locations of osteopontin and CD44 in kidney were detected by immunohistochemistry. The relationship of the osteopontin and CD44 expressions to calculus crystal depositon time dependently were analyzed. [Results] The expressions of osteopontin and CD44 were increasing with the deposition of calculus crystal time dependently before the day 14, and then decreasing. The expression of osteopontin kept the positive correlation to CD44 (P<0.05). [Conclusions] Under the hyperoxaluria stimulating,the expressions of osteopontin and CD44 were related to the deposition of calculus crystal and urinary stone formation, which might be the source of Randall plaque in rat with hyperoxaluria.
