[目的]探讨神经生长因子(NGF)对大鼠脑缺血再灌注后Fas-L和p-PKB表达的影响,为临床应用NGF治疗缺血性脑血管病提供依据。[方法]健康成年SD大鼠55只,随机分为假手术组(n=5);对照组(n=30),按缺血2 h再灌注2 h、6 h、12 h、24 h、48 h、72 h再分为6组,每组5只;NGF组(第8f和8p组,分别为缺血2 h再灌注6 h和再灌注72 h注射NGF,每组5只)和生理盐水组(第9f和9p组,分别为缺血2 h再灌注6 h和再灌注72 h注射生理盐水,每组5只)。制备MCAO模型。观察各组动物神经功能缺损评分和脑内Fas-L和p-PKB表达。[结果]NGF组与生理盐水组比较,神经功能缺损程度(1.400±0.6253,2.300±0.3650),(2.200±0.6903,3.100±0.0038)均明显减轻,差异有显著性意义(P<0.01)。与缺血再灌注组相比,NGF干预组Fas-L蛋白表达
(18.6600±12.2584)减少,p-PKB表达(66.6600±12.3353)增多,差异有显著性意义(P<0.01)。[结论]肌注NGF对脑缺血再灌注有明显的保护作用。 |

Abstract: |
[Objective]To observe the effect of nerve growth factor on expression of p-PKB and Fas-L gene protein after reperfusion in post cerebral ischemia rats,to provide the theoretical basis for clinical therapy of ischemia cerebrovascular disease. [Methods]Fifty-five adult SD rats were randomly divided into eleven groups with five in each,including sham group,6 control groups which according to ischemia time were divided into 2 h,6 h,12 h,24 h,48 h and 72 h ones,NGF groups including 8f one,NGF injected after 6 h reperfusing,and 8p group,NGF injected after 72 h reperfusing,and saline groups including 9f,saline injected after 6 h reperfusing and 9p group,saline injected after 72 h reperfusing. MCAO model was set up. Zea long 5 mark system standard grades was observed in each group of rats for nerve function appraisal and the expression of Fas-L and p-PKB protein was examined.[Results]The nerve function grade,compared with normal saline group (2.200±0.6903,3.100±0.0038),disfunction of NGF group (1.400±0.6253,2.300±0.3650) were obviously alleviated (P<0.01);compared with ischemia and reperfusion rats,NGF given in the early stage of ischemia could increase the expression of p-PKB protein(66.6600±12.3353) and reduce the expression of Fas-L protein (18.6600±12.2584,P<0.01). [Conclusion]Injecting NGF has obviously protective function in the ischemia- reperfusion rats. |
