摘要: |
[目的]观察NF-κB抑制剂吡咯烷二硫代氨基甲酸(pyrrolidine dithiocarbamate,PDTC)对顺铂毒性的影响。[方法]SD大鼠40只,随机分为4组,对照组和PDTC组各10只连续用药13 d,于第14天处死;顺铂组和PDTC-顺铂联合用药组各10只连续给药5 d后处死;测定体重和肾重、血肌酐、血尿素氮和白细胞数,用放射免疫法检测血清及肾组织匀浆TNF-α、IL-1含量。[结果]顺铂组大鼠肾系数增大为4.9440±0.5456,BUN升高至(61.073±44.782)mg/dL,血Cr增高至(9.743 ±3.821)mg/dL,肾组织匀浆IL-1含量升高;顺铂合用PDTC组的大鼠肾系数为4.4517±0.3658,BUN为(59.575±27.914)mg/dL,血Cr为(8.322±2.855)mg/dL,肾组织匀浆IL-1含量近正常。两组间肾IL-1含量比较,差异有显著性意义(P<0.05)。[结论]PDTC不能明显降低顺铂引起的肾毒性。 |

[Objective]To explore the effects of NF-κB inhibitor pyrrolidine dithiocarbamate (PDTC) on nephrotoxicity of cisplatin.[Methods]SD rats of 40 were divided into 4 groups.The rats of control group were daily injected with saline and those of PDTC group with PDTC for 13 days,and killed on day 14th.The rats of groups were daily injected with cisplatin and those with cisplatin+PDTC,and then were killed on day 5th.Weights of body and kidneys,blood urea nitrogen(BUN)and creatinine content of serum and white blood cell number of peripheral blood were measured,while TNF-αand IL-1 of serum and renal homogenate were measured with radioimmunoassay.[Results]In the rats injected with cisplatin,renal index was 4.9440±0.5456,BUN was (61.073±44.782) mg/dL,creatinine content of serum was (9.743 ±3.821)mg/dL,IL-1 concentration of renal homogenate was higher therefore in rats injected with cisplatin and PDTC,renal index was 4.4517±0.3658,BUN was (59.575±27.914) mg/dL,blood Cr was (8.322±2.855) mg/dL,IL-1 concentration of renal homogenate was normal. The difference between tow groups was significant statistically on IL-1 concentration of renal homogenate(P<0.05).[Conclusions]PDTC could not alleviate nephrotoxicity from cisplatin. |
