低能体外冲击波对大鼠成骨细胞增殖分化的影响

摘要:
[目的]探讨低能体外冲击波(ESW)对体外培养鼠成骨细胞(ROB)增殖、成骨分化的作用及其细胞内信号转导。[方法]每次取6只大乳鼠,取颅盖骨细胞分2瓶培养至第3代备用。取培养第3代ROB,用0.18 mJ/mm2 低能ESW不同次数(0,30,60,90,120,150次)刺激ROB,用细胞计数、MTT和流式细胞法检测ROB增殖状况,用酶标仪检测碱性磷酸酶(ALP)活性,用免疫组化检测I型胶原表达,观察ROB成骨分化,分析ESW对ROB的影响;然后,选择适当的ESW刺激(120次),并加入PKC抑制剂H7或者p38MAPK抑制剂SB203580。观察上述ROB增殖、分化及p38MAPK磷酸化激活状况变化。[结果]ESW0.18 mJ/mm2 刺激60~150次可显著促进体外培养ROB细胞增殖和成骨分化(与对照组比较,P<0.05);以120次刺激促进ROB增殖分化作用最强。PKC抑制剂H7和p38MAPK抑制剂SB203580都能明显抑制120次ESW的这一作用,PKC抑制剂H7能明显抑制ESW作用ROB后磷酸化激活p38MAPK的作用。[结论]适当的ESW应力刺激可能会促进体外培养ROB增殖和成骨分化,PKC和p38MAPK可能都参与此过程的细胞内信号转导。

Abstract:
[Objective]To investigate the effect of low density extracorporeal shock wave (ESW) on proliferation and differentiation of rat osteoblasts (ROB).[Methods]ESWs on 0.18 mJ/mm2 with different times (0,30,60,90,120,150 times),stimulate ROBs cultured from bone of newborn rats.The condition of cell proliferation was observed by cell counting,MTT and flow cytometry,cell differentiation by measuring ALP activity and type I collagen expression by immunohistochemistry.Then,ESW on 0.18 mJ/mm2 with 120 times was selected to stimulate cultured ROB and activity of p38MAPK was detected by Western blot.Furthermore,H7 as PKC inhibitor and SB203580 as p38MAPK inhibitor are employed to study PKC and p38MAPK's roles in the signal transduction.[Results]ESW stimulation on 0.18 mJ/mm2 with different times (60~150) might improve proliferation and differentiation of ROB cultured .The ESW with 120 times could gave the strongest.effect.Both of H7 and SB203580 can downregulate effect of ESW significantly,and H7can significantly inhibit the activity of p38MAPK stimulated by ESW.[Conclusion]Suitable ESW stimulation might improve proliferation and osteogenesis of cultured ROB,and both PKC and p38MAPK might contribute to the intracellular signal transduction in the process.
