低分子肝素对肾小球系膜细胞产生细胞外基质的影响

摘要:
[目的]了解不同葡萄糖浓度环境下,低分子肝素对人系膜细胞表达纤维连接蛋白(FN)和IV型胶原作用。[方法]培养人肾小球系膜细胞,分为正常葡萄糖组、高葡萄糖组及加低分子肝素组,培养24、48 h收集细胞上清液,用酶联免疫法检测FN和IV型胶原。[结果]高糖组24和48 h FN分别为(1.552±0.546)ng/mL和(2.547±0.968)ng/mL 明显高于正常糖组(0.777±0.076)ng/mL和(1.180±0.091)ng/mL(P<0.01);IV型胶原分别为(64.577±5.543)ng/mL和(80.140±6.605)ng/mL,明显高于正常糖组(42.468±5.615)ng/mL和(46.368±5.714)ng/mL(P<0.01)。在低分子肝素1 ng/mL 的作用下,高糖组的FN24 h及FN48 h分别降低为(1.070±0.349)ng/mL及(1.918±0.074)ng/mL,IV型胶原24 h及IV型胶原48 h分别降低至(41.993±7.470)ng/mL及(40.490±4.377)ng/mL。低分子肝素提高至10 ng/mL时,高糖组的FN24 h及FN48 h分别降低为(0.658±0.071)ng/mL及(0.280±0.035)ng/mL,IV型胶原24 h及IV型胶原48 h分别降低至(34.717±4.505)ng/mL及(27.902±5.325)ng/mL(P<0.01)。[结论]低分子肝素可抑制高糖刺激的系膜细胞表达FN、Ⅳ型胶原。

Abstract:
[Objective]To observe the effect of glucose concentration,low molecular weight heparin on human mesangial cells to produce FN and collagen Ⅳ.[Methods]Human mesangial cells were cultured.Then all cultured cells were divided into normal glucose group,high glucose group and experimental groups .Human mesangial cells were incubated with different glucose (5 mmol/L or 25 mmol/L) with or without different concentrations of low molecular weight heparin for 48 hours.Type Ⅳcollagen and fibronectin ( FN) levels were assessed by ELISA.[Results]Human glomerular mesangial cells in different glucose expressed FN and type Ⅳ collagen.Low molecular weight heparin decreased the level of cell culture supernatant of FN and type Ⅳ collagen in high glucose.The levels of FN in 25 mmol/L glucose at 24 or 48 hours being (1.552±0.546) ng/mL and (2.547±0.968) ng/mL were significantly higher than those in 5.0 mmol/L glucose being (0.777±0.076) ng/mL and (1.180±0.091) ng/mL (P<0.01).The levels type Ⅳ collagen in 25 mmol/L glucose at 24 or 48 hours being (64.577±5.543) ng/mL and (80.140±6.605) ng/mL were significantly higher than those in 5.0 mmol/L glucose being (42.468±5.615) ng/mL and (46.368±5.714) ng/mL (P<0.01).In the 25 mmol/L glucose group with low molecular weight heparin (1 mg/L),FN24 h and FN48 h decreased to (1.070±0.349) ng/mL and (1.918±0.074) ng/mL respectively,and the type Ⅳ collagen24 h type and Ⅳ collagen48 h decreased to (41.993±7.470) ng/mL and (40.490±4.377)ng/mL respectively. In the 25 mmol/L glucose group with low molecular weight heparin (10 mg/L),FN24 h and FN48 h decreased to (0.658±0.071) ng/mL and (0.280±0.035) ng/mL respectively,and the type Ⅳ collagen24 h or type Ⅳ collagen48 h decreased to (34.717±4.505)ng/mL and (27.902±5.325) ng/mL respectively(P<0.01).[Conclusion]Low molecular weight heparin may be decrease FN and type Ⅳ collagen expressions of cultured mesangial cell by high glucose stimulation.
