甘正复方对非酒精性脂肪肝大鼠PPARγ和CYP2E1表达的影响

摘要:
[目的]探讨甘正复方对非酒精性脂肪肝大鼠的疗效和作用途径。 [方法]SD大鼠随机分为正常组、模型组和治疗组。采用高脂饮食脂肪肝模型,治疗组用甘正复方灌胃,实验周期16周。检测指标:血清ALT、AST、TNFα和脂联素水平;肝组织脂肪变性及炎症活动度计分;肝组织CYP2E1、PPARγ蛋白和mRNA的表达。 [结果]模型组血清ALT、AST增加,治疗组较模型组血清ALT、AST下降(P均<0.01);病理学显示模型组脂肪变性及炎症程度积分高于正常组,治疗组低于模型组(P<0.01);模型组TNFα、CYP2E1较正常组明显增高,治疗组明显低于模型组(P<0.01)。模型组血清脂联素、PPARγ表达减少(P<0.01);治疗组肝组织表达明显高于模型组(P均<0.01)。[结论]甘正复方通过调控PPARγ、脂联素和CYP2E1的表达等多种途径有效地治疗脂肪肝。

Abstract:
[Objective]To investigate the effects and the mechanism of Ganzhengfufang on nonalcoholic fatty liver disease (NAFLD).[Methods]SD rats were divided into control group,model group and treatment group by random.High fat diet model was employed.Treatment group was PO dosed with Ganzhengfufang while control group was fed with water.The whole duration of the study was 16 weeks.Serum ALT,AST,TNFα and Adiponectin,the histology of liver steatosis and inflammation score,the expression of CYP2E1,PPARγ and mRNA in liver tissue were tested.[Results]ALT and AST were increased in model group,while reduced in treatment group (P<0.01).The steatosis and inflammation score were higher in model group,but were lower in the treatment group (P<0.01).TNFα,CYP2E1 were increased in model group but decreased in treatment group (P<0.01).The expression of adiponectin and PPARγ decreased in model group,increased in treatment group.[Conclusion]Ganzhengfufang can have effect on the NAFLD rat by means of regulating PPARγ,adiponectin and CYP2E1.
