石蜡半薄切片制作透射电镜的方法

摘要:
[目的]介绍石蜡半薄切片转制透射电镜的制样方法。[方法]对石蜡半薄切片进行脱蜡、水化、锇酸固定、缓冲液漂洗、梯度酒精脱水、环氧树脂包埋、超薄切片机切片、透射电镜下观察。[结果]组织细胞的超微结构保存完好,足以满足病理诊断之用。[结论]用石蜡半薄切片转制电镜比用石蜡组织块转制电镜方法简单,快速易行。

Abstract:
[Objective] To introduce sample preparation methods of TEM using semi-thin paraffin sections. [Methods] The semi-thin paraffin section was processed with dewaxing, hydration, osmium tetroxide fixed, buffer rinsing, gradient alcohol dehydration, epoxy resin embedded and ultra-thin slice. Then the sections were observed under the Transmission Electron Microscopy. [Results] The ultra-structure of histo-cyte remained well. It was sufficient to meet the requirement of pathological diagnosis. [Conclusion] Compared with paraffin tissue samples, using semi-thin paraffin sections are more efficiency.
