摘要:
介绍应用siRNA表达框架对原代培养的细胞进行RNA干扰。RNA干扰是由双链RNA介导的特异同源靶基因转录后沉默的过程,它已经广泛应用到基因功能研究、基因表达调控机制和抗病毒感染等热门领域,在哺乳动物的原代细胞中进行RNA干扰的研究为功能基因研究提供了重要意义。siRNA表达框架与脂质体的转染率直接影响RNA干扰的效果,本文介绍了如何设计和构建siRNA表达框架,筛选最佳的靶位点。

Abstract:
To introduce the process of initiating RNA interference mediated by siRNA expression cassettes in primary cell. RNA interference (RNAi) is a highly specific form of posttranscriptional gene silencing mediated by double-stranded RNA. RNAi has rapidly become one of the most powerful functional genomics tools recently and has the potential to be used therapeutically. The degree of success in silencing a specific target RNA in primary cultured mammalian cells is often dependent on siRNA or plasmid transfection efficiency. This article describes how to design and synthesize siRNA expression cassettes and, additionally, find the optimal target site for RNAi-mediated gene silencing.
