胃癌细胞SGC7901/ADR耐药相关蛋白质分子的差异展示

摘要:
[目的]应用蛋白质组学技术寻找新的胃癌多药耐药相关蛋白分子,探讨胃癌多药耐药机制。[方法]以胃癌细胞SGC7901和阿霉素诱导的耐药胃癌细胞SGC7901/ADR为研究对象,用固相pH梯度等点聚焦二维聚丙烯酰胺电泳技术展示两种细胞表达的全蛋白,凝胶考马斯亮蓝染色, Image Master 2D Melanie5.0凝胶分析软件分析并比较差异表达的蛋白分子。[结果] SGC7901中检测到414个蛋白点,SGC7901/ADR检测到553个蛋白点,两者匹配点386个,差异点71个,其中SGC7901/ADR蛋白质表达明显上调蛋白26个点,明显下调蛋白点45个。[结论] SGC7901/ADR细胞中差异蛋白分子可能与胃癌阿霉素耐药机制有关。

Abstract:
[Objective] To find for new multidrug-resistant proteins and discuss the new multidrug-resistant mechanism in gastric cancer cells. [Methods] Immobilized pH gradient-based two-dimentional gel electrophoresis was used to show the proteins in gastric cancer cells SGC7901 and adriamycin-resistant SGC7901 cells (SGC7901/ADR). The gels were used to stain the protein with commassie blue. ImageMaster 2D Melanie 5.0 software was used to analyze the differential expression proteins. [Results] In SGC7901 gel and SGC7901/ADR gel 414 and 553 protein spots were identified respectively and 386 protein spots were matched. There were 71 obviously differential expressional protein spots, 26 spots higher and 45 lower in quantity in SGC7901/ADR gels. [Conclusion] The differential proteins in SGC7901/ADR may be related to the adriamycin-resistant mechanism.
