摘要: |
[目的]探讨邻苯二甲酸(2-乙基已基)(Diethylhexyl phthalate,DEHP)对小鼠雄性生殖系统的毒性作用机制。[方法]昆明种雄性小鼠(20±2)g,40只,随机分为4组,每组10只。经食饵自然给食染毒,连续4周后处死。观察小鼠体重及睾丸变化,测定不同剂量DEHP染毒小鼠的血液、睾丸中谷胱甘肽过氧化物酶(GPX)活性、一氧化氮(NO)和过氧化氢(H2O2)含量。[结果]DEHP染毒后,随着染毒剂量增加,小鼠体重逐渐减少(P<0.01),高剂量组睾丸脏器系数显著下降(P<0.01)。光镜下可见高剂量组睾丸组织有明显损伤。与对照组相比,DEHP中、高剂量暴露组小鼠血液及睾丸中GPX活性降低(P<0.01);H2O2 含量增加(P<0.05,P<0.01);睾丸中NO含量降低,尤其是高剂量组与对照组差别明显(P<0.05,P<0.01)。[结论]DEHP对小鼠雄性生殖系统具有明显的毒性作用,其作用机制可能与氧化应激反应和NO含量降低有关。 |

[Objective] To detect the mechanism of male preconception injuries induced by diethylhexyl phthalate (DEHP) in mice.[Methods] Forty of Kumming mice (20±2)g were divided into 4 groups randomly,every group including 10 mice. The animals were exposed to DEHP by food for 4 weeks.At the end of the fourth week,they were terminated.Then calculated the weight and coeffcient of testis/body. GPX、NO、H2O2 of blood and testis were examined.[Results] With the increase of DEHP ,the weight of mice and the coeffcient of testis/body with the high dose group decreased significantly (P<0.01). There were evident testes tissue damage in high dose group with examination by light microscope. Comparing with the control group,GPX activity in the blood and testes decreased dramatically (P<0.01), while H2O2 content increased significantly (P<0.05,P<0.01); in modprote and high dose qroups NO content decreased dramatically in testes,especially in high dose of DEHP (P<0.05).[Conclusion] DEHP has significant male preconception injuries effect to mice. The possible mechanisms are oxidative stress injuries and NO content decrease. |
