[目的]研究基因重组白眉蝮蛇去整合素Adinbitor对SSMC7721 细胞增殖、凋亡及侵袭的影响。[方法] 用Hoechst33258试剂盒染色,观察Adinbitor对细胞形态的影响;Western-blotting 检测浓度梯度Adinbitor作用下Caspase-3表达情况,分光光度法考察Caspase-3活化程度;MTT法检测Adinbitor对细胞增殖的作用,Transwell及Matrigel检测其对细胞侵袭的影响。[结果]Hoechst染色显示Adinbitor可明显促进细胞凋亡。在Adinbitor作用下,胞浆Caspase-3含量减少,活化程度增高,活化倍数2.24~3.85。Adinbitor可明显抑制SSMC7721细胞增殖,IC50为0.296 μmol,也可抑制细胞侵袭能力。[结论] 基因重组Adinbitor可明显抑制SSMC7721细胞的增殖及侵袭,并且可通过活化Caspase-3促进细胞凋亡。 |

[Objective] To explore the effects of a disintegrin, Adinbitor on proliferation, apoptosis and invasion of human hepatoma cells.[Methods] MTT assay was used to detect the effect of Adinbitor on proliferation. SSMC7721 cells were incubated with different concentrations of Adinbitor and stained by Fluorescence dye Hoechst 33258 to observe apoptosis .The amount of Caspase-3 was detected by Western blotting. Spectrophotometric method was selected to explore the activity of Caspase-3. Transwell and matrigel were used to observe the effect of Adinbitor on cell invasion. [Results] Adinbitor could inhibit cell proliferation and enhance cell apoptosis. The amount of Caspase-3 in cytoplasm were decreased. The activatory fold of Caspase-3 was from 2.24 to 3.85. The inhibition IC50 of Adinbitor on cell proliferation was 0.296 μmol . The amounts of invasive cells were decreased significantly, versus control group, P<0.05. [Conclusion] Adinbitor could inhibit SSMC7721 cell proliferation and invasion and it could induce apoptosis by activation of Caspase-3. |
