[目的]探讨骨髓基质干细胞(bone marrow stromal cells,MSCs)不同途径移植后在急性局灶性脑缺血疾病治疗中的作用及其机制。
[方法] 利用线拴法制作SD大鼠大脑中动脉缺血模型(MCAO)(n=80),随机均分为:缺血自然恢复组(A组),缺血+磷酸盐缓冲液(PBS)对照组(B组),缺血+颈内动脉MSCs组(C组)和缺血+尾静脉MSCs组(D组),分别通过颈内动脉、尾静脉移植相同数量MSCs。再灌注24 h后,分别观察神经行为学评分,脑梗死体积,尼氏小体平均灰度,凋亡相关基因Bcl-2,Bax及血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达。[结果]通过对神经行为学评分,脑梗死体积和病理形态学(尼氏小体、Bcl-2、Bax免疫组化以及VEGF的表达)分析,C组最终的各种指标分析比较均要优于其他3组,之间的差异有显著性意义(P<0.05)。[结论] 颈内动脉移植MSCs治疗缺血性脑损伤的疗效明显优于尾静脉移植相同数量的MSCs。 |

Abstract: |
[Objective] To investigate the effect and mechanism of bone marrow stromal cells (MSCs) transplantation for treating cerebral ischemia in rats. [Methods] Rat middle cerebral artery occlusion (MCAO) model was established. A total of 80 SD rats were divided into four group, MSCs ( 2×106 ) labeled was transferred into every group, through external carotid artery 24 h after stroke. Behavioral tests were performed before and after MCAO. Immunohistochemistry was used to identify MSCs and expression of neurotrophic factors. [Results] The transferred MSCs survived, migrated and producing neurotrophic factors, such as VEGF after the transplantation. Compared with other three groups, the function of nervous system was significantly improved ( P<0.05 ). And, the therapeutic effect of the different administration channel, C group was better than the D group. [Conclusions] The therapy of MSCs transferring into rat brain after cerebral ischemia is effective. The neurotrophic factors produced by MSCs may explain the protective mechanism. |
