[目的]探讨复方白花前胡液对拟缺血诱导的神经细胞凋亡的影响及其作用机制。[方法]培养的细胞随机分为空白对照组、模型组、复方白花前胡液大剂量组、复方白花前胡液中剂量组,每组10孔。将培养的神经元细胞采用缺氧罐和无糖Earle's液进行缺氧/缺糖处理3 h,再复氧复糖24 h后,显微镜下观察培养的海马神经元细胞的形态,流式细胞仪检测细胞凋亡率,并分别检测乳酸脱氢酶含量及细胞游离钙的含量。[结果] 1.复方白花前胡液中、高浓度实验组细胞凋亡率分别下降至(21.4±3.9)%、(23.6±2.8)%,与模型组相比差异有显著性意义( P < 0. 01 )。2.各复方白花前胡液组在不同时点LDH释放率与模型组相比降低明显,差异有显著性意义( P< 0.05 )。 3.复方白花前胡液实验组与模型组比较缺氧/缺糖培养3 h,复氧复糖24 h胞内Ca2+ 浓度下降,差异有显著性意义( P < 0.05 )。[结论]复方白花前胡液能使神经细胞具有抗缺氧/缺糖损伤作用。 |

[Objective]To investigate the effect of FFBHQHY (Chinese Medicine) on the damage of nerve cells caused by hypoxia/hypoglycemia.[Methdos] Cultured cells were randomly divided into normal group, model group, High-dose FFBHQHY group, dose FFBHQHY group, with 10 holes in each group.Cultivated the hippocampal cells of fetal rats then left them in the situation of hypoxia/ hypoglycemia.The shapes of the propidium iodide cells were observed. The change of apoptosis percentage caused by hypoxia/hypoglycemia was examined through flow cytometry(FCM). [Results] 1.The apoptosis ratios in FFBHQHY groups were ( 21.4 ± 3.9 )%, ( 23.6 ± 2.8 )% ( P< 0.01).2.Compared with the normal group, FFBHQHY group, LDH decreased significantly ( P<0.01 or P<0.05 ).3.The free Ca2+ decreased notably in FFBHQHY compared with that in the control group ( P< 0.05 ). [Conclusion] FFBHQHY could resist hypoxia/hypoglycemia. |
