[目的]发现炎症内皮释放的一种新的趋化因子Fkn与其受体CX3CR1结合后,可导致血管内皮细胞损伤,并使表达该受体的单核细胞、淋巴细胞等产生方向性迁移信号,向血管内皮细胞趋化、黏附,并能穿过血管向血管外特定部位迁移,从而促进冠状动脉粥样硬化的形成。本实验探讨CX3CR1基因多态性和经过冠状动脉造影确诊为冠心病患者之间的关系,以分析CX3CR1基因多态性是否对冠心病的发病有影响。[方法]应用聚合酶链反应—限制性片段长度多态性(PCR-RFLP)技术分别对108例冠心病患者和80例健康者的CX3CR1基因进行多态性检测,分别计算冠心病组和健康对照组的3种基因型频率。[结果]CX3CR1的I249等位基因的基因频率在冠心病组和健康对照组上有显著差别;M280等位基因和I249等位基因的频率结合起来,得出调整后的OR值分别是0.49和0.43。[结论]CX3CR1的I249等位基因在冠心病组和健康对照组的基因频率上差别有显著性意义;M280、I249等位基因和冠心病发生的减少有关;其中等位基因I249起执行和保护的作用,能显著减少冠心病的发病率。 |

[Objective]We further test the hypothesis by analyzing the association of CX3CR1 polymorphism with angiographically defined atherosclerosis.[Methods]Genotyping of the CX3CR1- I249 polymorphism was performed in a cohort of 188 yellow individuals who undergone cardiaccatheterization (n=108 with and n=80 without cardiac artery disease, respectively). [Results]The frequencies of the I249 and M280 polymorphisms showed no deviation from Hardy-Weinberg equilibrium. However, a statistically significant difference in genotype frequencies was observed in cases compared with controls. The adjusted odds ratios associated with the presence of the M280 and I249 alleles were 0.49 and 0.43.[Conclusions]CX3CR1 I249 heterozygosity is associated with a markedly reduced risk of acute coronary events, independent of established acquired coronary risk factors. The CX3CR1 I249 allele is associated with decreased risk of cardiac artery disease. This suggests that CX3CR1 may be involved in the pathogenesis of cardiac artery disease. |
