[目的]研究乌苏里藜芦生物碱对异丙肾上腺素诱导的小鼠心肌肥大的保护作用,并对其作用机制进行初步探讨。[方法]小鼠尾静脉注射乌苏里藜芦生物碱6 h后,给予异丙肾上腺素(3 mg/kg)连续5 d。末次给药24 h后处死小鼠,称取小鼠全心重及左心室(含室间隔)重量,取左心室匀浆测定SOD、MDA、Ca2+ -ATP活性,HE染色和透射电子显微镜观察心肌结构。[结果]高剂量和中剂量乌苏里藜芦生物碱均可降低心脏指数,使心肌MDA水平降低 [高剂量组(2.39±0.48)nmol·mg-1 prot,与模型组(3.56±1.08)nmol·mg-1 prot比较,P<0.01],SOD [高剂量组(131.32±20.07)
nmol·mg-1 prot,与模型组(107.43±7.86)nmol·mg-1 prot比较,P<0.05]和Ca2+ -ATP酶活性升高 [高剂量组(5.78±0.51)μmolpi·mgprot -1·h-1,与模型组(4.06±0.38)μmolpi·mgprot-1·h-1 比较,P<0.01],病理切片和透射电镜观察证明心肌损害有明显改善。[结论]乌苏里藜芦生物碱在15~45 μg/kg呈剂量依赖性抑制异丙肾上腺素诱导的小鼠心肌肥大,这一作用可能与其抗氧化、激活心肌Ca2+ -ATP酶活性有关。 |

[Objective]To investigate the protective effect of Veratrum nigrum L. Var.ussurience Nakai alkaloids (VnA) on cardiac hypertrophy induced by Isoproterenol (ISO) in mices.[Methods]Therapy group were given VnA i.v. 6 hours before ISO treatment. Then we invertigated the cardiac weight index and the MDA content, the activity of SOD and Ca2+ -ATPase in myocardium, myocardiac morphological alterations were observed by pathological section and transmission electron microscope. [Results]VnA of the high dose and middle dose were effective to decrease the cardiac weight index and MDA content [the high dose group is (2.39±0.48) nmol·mg-1 prot VS the model group (3.56±1.08) nmol·mg-1 prot, P<0.01],improve the activity of SOD [the high dose group is (131.32±20.07) nmol·mg-1 prot,VS the model group (107.43±7.86) nmol·mg-1 prot, P<0.05)] and Ca2+ -ATPase in myocardium [the high dose group is (5.78±0.51) μmolpi·mgprot-1·h-1, VS the model group (4.06±0.38)μmolpi·mgprot -1·h-1, P<0.01]; VnA improved the left ventricular tissue morphological alteration.[Conclusion]VnA possess protective effects against Isoproterenol that induced cardiac hypertrophy in mices and dose-depent from 15~45 μg/kg. This may be related to reducing the oxidative stress,activatingCa2+ -ATPase of myocardial cells. |
