[目的]对槐花米中槲皮素传统的制备方法进行改进,从而提高槲皮素的产率和提取效率。[方法]利用碱提取-酸沉淀法从槐花米中提取芦丁粗品,采用:①间接酸水解法(传统方法),将粗制芦丁通过重结晶纯化得到精制芦丁,再用酸水解制得槲皮素;②直接酸水解法(改进方法),将粗制芦丁的重结晶滤液直接加酸水解制得槲皮素;并用HPLC等技术对两种方法所得产品与槲皮素标准品进行鉴定和分析。[结果]改进方法制备的槲皮素与传统方法相比,二者纯度均与标准品接近,但前者反应时间至少缩短120 min,产率却提高19.45%。[结论]改进方法显著地提高了槲皮素的提取产率和效率,为槲皮素的实验室制备和工业生产提供借鉴。 |

[Objective]Improved the preparation method of quercetin to increase its yield and efficiency.[Methods]Extracted rutin from the bud of Sophora japonica by alkali extraction-acid precipitation method, and then prepared quercetin by :①recrystalling rutin and then hydrolyzing indirectly the solution of rutin crystal with sulfuric acid (conventional method); ②hydrolyzing directly the recrystallization filtrate of rutin with sulfuric acid (improved method), identifing the product by HPLC.[Results]There was no difference between two methods in purity, whereas the reaction time became shorter 120 min at least and the yield was raised 19.45% in the improved method.[Conclusions]Compared with the conventional method, the improved one is simpler, as well as the extraction efficiency and yield of quercetin was significantly increased, and which would be used as reference to laboratory-scale preparation and industrial production. |
