[目的] 从细胞水平研究维生素E琥珀酸酯(VES)对顺铂(CP)引起的前列腺癌(LNCaP)细胞的增殖抑制和凋亡诱导作用,并分析其对前列腺特异性抗原(PSA)表达的影响。[方法]VES、CP及二者联合分别作用LNCaP 24 h或48 h,采用MTT法测定其对细胞生长的抑制作用,用荧光染色和透射电镜等形态学方法检测细胞凋亡,并用酶免疫法检测前列腺癌细胞特异性抗原(PSA)的表达情况。[结果]CP、VES和二者合用的IC50 分别为1.92 mg·L-1、16.7 mg·L-1和1.02 mg·L-1;CP 4 mg·L-1、VES 25 mg·L-1和二者合用的凋亡率分别为37.2 %、57.4%和77.8%;VES 可以加强CP抑制LNCaP细胞分泌PSA的作用。[结论]VES可增强CP抗LNCaP细胞的作用。 |

[Objective]To investigate the growth inhibition and apoptosis against cisplatin-induced of vitamin E succinate on human prostate cancer cells and analyze effects of VES on levels of secreted prostate-specific antigen(PSA) in the cells.[Methods]Human prostate cancer cells LNCaP were treated 24 h or 48 h with CP alone, VES alone or a combination of both. The inhibitory effect was measured with MTT method and the cell apoptosis were assessed with the ethidium bromide fluorescence and electron microscope, PSA of LNCaP were examined with ELISA.[Results]IC50of CP, VES or a combination of both were 1.92, 16.7 and 1.02 mg·L-1, respectively, the apoptotic rate was 37.2 %, 57.4% and 77.8%, respectively, after treated with CP 4 mg·L-1, VES 25 mg·L-1and both of them. VES can decrease the PSA level of LNCaP cells and the effect was more obvious after treatment with VES in combination with CP.[Conclusions]VES can enhance the efficacy of anti-LNCaP cells by CP. |
