[目的]探讨榄香烯、丝裂霉素C、NDV对HCa-F和L615瘤细胞生物学性状的影响。[方法]将HCa-F、L615细胞分别用50 μg/mL或100 μg/mL榄香烯(elemene,E)、50 μg/mL丝裂霉素 (mitomycin C,MMC)处理,未处理的细胞为对照组,观察细胞存活率;电镜观察超微结构的改变;将HCa-F、L615瘤细胞处理后,流式细胞仪观察凋亡和细胞周期的变化。[结果]HCa-F细胞在50 μg/mL E处理的0~6 h,存活率从(99.8±0.2)%变为(30.8±5.8)%,100 μg/mL E处理2 h时已无存活的HCa-F细胞(P<0.01);L615细胞经50 μg/mL E处理2 h,存活率即为0(P<0.01)。在HCa-F细胞内可见染色体边集、核固缩及凋亡小体;L615细胞各组凋亡细胞率分别为(17.70±0.85)%,(32.72±1.32)%,(55.13±1.09)%(P<0.01),而HCa-F细胞凋亡率分别为(2.15±0.08)%、(3.76±0.34)%、(5.60±0.57)%、(8.00±1.20)%(P<0.01);E单独处理L615细胞时其S期细胞百分率增加为(45.6±8.9)%(P<0.01),但单用MMC或E-MMC复合处理却使L615 细胞的S期比率减少为0(P<0.01)。[结论]榄香烯与丝裂霉素C在细胞毒效应方面存在协同作用,使肿瘤细胞阻滞在S期,减少进入G2-M期的细胞数目。 |

[Objective]To inrestigate the chility of elemene,mitomycin C and Newcastle disease virus to suppress cell proliferate and survival,and induce apoptosis of HCa-F and L615 tumor cells in culture.[Methods]HCa-F and L615 cell survival ratse were estmated by estmated by cells which stained with trypan bule following the treatment of elemene (E) in doses of 50 μg/mL and 100 μg/mL. The ultramicrostructure of calls by electron microscope after the treatment of and their control HCa-F calls reacted for 1 hour with E, mitomincin C(MMC), Newcastle disease virus(NDV)or the comlination of each other. Apoptosis rate and cell cycle of the tumes cells were studied by folw cytomety after treatment of fresh Hca-F and L615 cells with mentioned drugs in singular or comlined each other.[Results]Survival rate of HCa-F is reduced from (99.8±0.2)% to (30.8±5.8)% during the cells were treated by 50 μg/mL of elemene.while HCa-F cells were treated with 100 μg/mL of elemene for 2 h,there is no survival HCa-F cell. A lot of morphological characteristics of apoptosis of HCa-F including compaction and margination of nuclear chromosome and apoptotic bodies were found in the treatment with elemene, MMC and NDV.The measured apoptotic cell rates of L615 cells were (17.70±0.85)%,(32.72±1.32)%,(55.13±1.09)% after treating for 1 h with E,M, and E-MMC respectively (P<0.01),the measured apoptotic cell rates of HCa-F cells were (2.15±0.08)%,(3.76±0.34)%,(5.60±0.57)%,(8.00±1.20)% after treatment with E,E-MMC,NDV, and NDV-E(P<0.01);Cell percentage of S phase rose to (45.6±8.9)%(P<0.01)after L615 cells treated with elemene singly,but pencentage of S phase was 0 % after treatment with MMC or E-MMC (45.6±8.9)%(P<0.01).[Conclusions]There is a synergistic cytotoxic effect with the comlinaton of elemene and mitomycin C.Changes of tumor cell ultramicrostructure were similar and the death modes of different tumor cells are not identical under treatment after each treatment. Elemene caused tumor cell arrest at S phase and resulted in the cell amount at G2-M phase reduced. |
