胃癌组织hMSH2基因突变的检出

摘要:
[目的]研究hMSH2基因表达及其突变在胃癌发生中的作用。 [方法]应用反转录—聚合酶链反应(RT-PCR)技术检测24例胃癌组织及15例癌旁组织的 hMSH2 mRNA表达情况。[结果]胃癌组的hMSH2 mRNA检出率为50%(12/24),高于癌旁胃黏膜组的20%(3/15);胃癌组织突变检出率为20.8%(5/24),高于癌 旁胃黏膜的0%(0/15);5例突变中,杂合性突变2例,纯合性突变3例。 在配对的15例中,仅胃癌组检出突变3例,均为纯合性突变。[结论]胃癌的发生伴随着hMSH2 基因的表达上调,这种表达上调可能与该基因突变有关。

Abstract:
[Objective] To study the roles of hMSH2 gene expression and mutation in gastric carcinogenesis. [Method] Reverse transcription polymerase chain reaction (RT-PCR) was used to examine the expressions and mutations of hMSH2 gene in 24 cases of gastric carcinoma tissues in comparison with those in 15 cases of their peri-carcinoma tissues. [Results] The detective rate of hMSH2 mRNA in gastric carcinoma tissues being 50% (12/24) was higher than that in peri-carcinoma tissues being 20% (3/15). The detective rate of hMSH2 gene mutation in gastric cancer tissues being 20.8% (5/24) was higher than that in peri-carcinoma tissues being 0% (0/15). In the 5 mutational cases, there were 2 heterozygous mutations and 3 homozygous mutations. And in the 15 paired cases, 3 homozygous mutations were only found in gastric carcinoma tissues. [Conclusion] The up-regulated hMSH2 mRNA expressions, which may related to the mutation of the hMSH2 gene, associated with gastric carcinogenesis.
